How well does the pipe cutter work? I used my scalpel last time and it was a real PITA.

At what temp will agar resolidify?

How long after getting off pressure do you wait before pouring?

Invest in a temperature gun and pour when gets down to about 120f. Can take quite a while for agar to cool after PC cycle.

Generally as soon as you can pour without the bottle burning your hand is a good temp.

I’m not sure when it solidifies but the window between pouring temp and when it starts to turn to sludge is very short.

Mine always starts getting thickened globs after it hits about 115f.  I like to let it cool to about 130f naturally in the PC then stick it in a 120f water bath until I'm ready to pour it.  If you stick it on a cool surface or anything that conducts heat away from the jar, you may get premature cooling/solidification at the bottom of the bottle.

If you have a sous vide machine it works wonders for agar. Set it to your desired temp(120-130) and put in your media right from the pressure cooker. Some have their own temperature sensor that tells you the temp of the water. As soon as the water is at your set temp(120-130) it's ready to go but you can leave it there for hours with no problem. Actually quickens the cool down process from the PC as well. Takes less than 30 minutes from the PC to pouring. Never had an issue.

That's if you have a sous vide though, just tossing that nugget out there as I've always happened to have one on hand vs a heat gun or fancy lab water bath which basically does the same thing.

Yeah I use my instant pot for agar so I can keep it warm, but that level of warm is still a bit too hot to handle well. I turn it off and wait a bit before pouring, but getting the sweet spot is still difficult.

Temp gun seems like a great $20 dollar investment so cheers!

Mostly curious about the nat plate. What causes like the different 'layers'?

A temp gun is absolutely a worthy investment. Aside from agar, I use it to make sure the area where I have things colonizing/fruiting isnt too warm, since I'm in an apartment and relying on a window AC. It's a quick way to verify temperature for anything at the snap of a finger. 10/10 would recommend.

At this point I do the same.
I microwave to heat it, shaking often- until it just starts to boil.
Needs a constant eye or it will be a mess haha.
I think that was Bods tek…
Let it rest for about 25/30 minutes.
I’ve had pretty good results in deli cups/containers that way.
One of these days I’ll get an IR thermometer to check things .

You guys like using the instant pot as a water bath?
My IP analog hits about 158 at the lowest (?)
Been thinking about doing that too.

Does it have a "Keep Warm" setting? It should, and if so, you can hit the keep warm button a couple of times until it says LOW. Then the temperature will keep at 135f(57c). So if you only fill it about 1/4 to 1/3 up the bottle and leave the lid off it makes a pretty good water bath. Some models have a sous vide function but mine doesn't. =/

I use a sous vide so I can set the temp exactly where I want it.  As an added bonus it can be used to pasteurize bags of substrate that require it.

I am with PBJ and the sous vide

Has become a tool I can't easily live without

I got it for pasteurizing but it works lovely for holding agar at pouring temp indefinitely, no more rushing to make the workflow fit the agar temp. Pour whenever you are ready, at your leisure

This just in:
1 gallon Ziploc bags are perfectly sized for two dozen 90mm plates.

That is all.

A few transfers from store bought lc. Original plates didn't look bad but im trying to learn so i transferred. How these lookin?

Tried my first clone tissue to agar two days ago from an extremely large pin that was in a pf jar. Nothing really going on there yet. Trying multiple things to see what works and what doesnt. Figure I'll learn a lot from failures. Had one plate with a perfect yellow circle on it so who knows what that is...other than in the garbage.
