Plates stored in doubled bagged ziploc freezer bags are pretty stable. Have you tried double bagging? I keep the outer bag inflated a bit to act as insulation. Single bagging works fine though.

I never double bag. I usually just toss a few wrapped plates into a gallon bag and leave it partially sealed. I thought they needed air.

Plenty of it in the bag. If you're leaving the bag open it might account for the drying.

I will definitely try bagging some and see how they hold!
Solid advise!

This stuff started showing up on several of my plates recently. Does it look like some kind of common contaminant?

We all have our struggles with bacteria. What’s your sterile procedure like? Looks like there’s a hole in it somewhere. Just gotta find it and seal it up.

Is that over or under the plate? Almost looks like it's growing between the bottom of the dish and the gel.

Not sterile enough it seems. I’m really new to working in a SAB, I’m sure it here’s a lot of room for improvement.

Mostly over, but now that you ask I shined a flashlight through it and it is wrapping around to the other side.

Looks like stuff people here build usually or just go to Harbor Freight for. Save money and DIY for some things -- but if you really like the quartz scalpel -- I'll personally keep my blade metal until it rusts through :hairmetal:.

The punches are cool though. D3 transfer style.

I’d probably remove the link. Easy enough to find and not one of ours. Always interesting to see what’s out there though.

This is a T1 plate. What do I have going on here? Weak myc with one strong sector? Or strong mold?

It all takes practice. How long do you PC your agar for? Cause if it was just like 15mins there's a chance they weren't sterile b4 you used them. This is also why you should let your plates sit blank for 2 weeks before you use them.

That could be it- I just started using the instantpot for agar instead of the proper pc. 20 minutes, and I think the instantpot is around 12psi. I thought this would be enough to sterilize liquids. I’ll experiment with making a batch that I won’t use right away with a longer cycle. Thanks!
