Re: AGAR ENVY! (Anything and All things agar!) [Re: CocaineBuffet]
    #28442036 -

My dive into agar work has been going well, so thanks to yall for pushing me in the right direction.

I'm still taking my time with transfers, slicing stronger and stronger looking growth from two varieties of cube.  I've been using a 3g potato, 2g agar, 1 cup water ratio and it's been successful.

I'll be ready for my first agar to grain soon.  So my question, should I use any specific recipe for my cups that I plan on transferring to grain?  Or even a water agar plate? Should I make a stiffer agar for easier mixing of slices with grain?

I'm still a couple weeks away from my final (hopefully) transfers.  I'll be making cups based on suggestions in a couple days.  Thanks always for yall's help.

This makes sense to me, seeing as the myc would be more evenly spread around the grain.  I'll have to convert to a smaller recipe, but I may just try it.

Anyone else do it this way?

Kept this plate around becauseI liked how it looks but at the same time it has been interesting to observe the growth and how slow it has been.

It's always known that contamination will run rife in days yet this plate is almost a month old and not filled yet.

Colder temperatures but cubensis myc transferred the same day has grown more.

You don't mix anything around on a plate like you do when you shake your grains and spawn to bulk

That's not my point, I've seen contamination fill a plate when it's enmesshed with cube mycelium for example.

I'm specifically referring to plates of agar hence posting in the agar thread.

Some results of "normal" LME Agar vs. Tea Agar. The Tea agar seems to really do a good job. I hope it still looks good after i transfer it back to LME Agar. Will still try to be patient and give it more time to grow out before transferring

Heres what think may be a successful cleanup with a tea agar sandwich

i would say my attempts at cleaning a culture with water agar failed unfortunately. Dont really trust what i see here. The agar seems to grow into the agar and also not uniform at all.

bit of an update with my first go working with agar, first pic is the plate i took the transfers from, they were taken from the leading edges at the time (albino A+ MSS ) the tranfers seem to be doing ok but would appreciate a heads up if anyone sees anything that should not be there , last 2 look the best to me (how many tranfers are ppl taking before going to grai - or just when it looks clean? - and what do ppl do with the tranfer plates -just leave them and clone if they pin invitro?

That's quite interesting - I'll have to give this a controlled test some time.

What’s everyone doing with mother/transfer agar dishes? This is my first grown and I have a lot of left over dishes and I’m still only on transfer one. Do you send them to grain or just toss them outside?

You can just leave em but you’ll end up with a wall of plates. I’ve been somewhat terrible but throw a lot away that are either sub par or transferred from and I don’t care anymore. To me once it’s used it’s used. It’ll never have that new plate smell anymore. I guess you could fridge em but there are better ways to hold on to things (slants in the fridge!).

Ya I kind of figured they would be good as garbage. I’ll hang on to them till I need the space I guess. Thanks.

All of my agar not destined for grain will eventually make it out to my compost bin. I keep hoping some take and I see cubes pop out when it rains. No luck yet, but I'll keep tossing them in there. Nothing I can think of better to do with them.
