If you have a real pressure cooker then definitely use it. You CAN use an instapot, but you have to increase time quite a bit. Idk stipe did a whole writeup on the science behind it.

But yeah if you’ve got a PC just use that. Way better.

according to this go for 150mins to really make sure it's clean

but I've found that makes the agar darken up a lot. maybe caramelizing the fructose in lme at that length?

pudin's instandpot tek says to go for 20 mins

I've only had contam in the cups that were intended to be contaminated, so it seems to work well at 150mins. I have a run going right now for 30 mins to see if the shorter time still works ok.

I use an instant pot for agar. I use 500ml media bottles, filled to 400ml. I vent for 15 and cycle for 45 min and then leave it on "keep warm" until it depressurizes and cools to the holding temp. (about 55c/130f). Using a proper PC is always a great choice, but an IP is a viable option.

Hoping this proves clean. Growth started to show yesterday. Still a little early for me to call the lc clean

It looks nice so far. What's the strain?

Anyone have thoughts on where to transfer from here? Or send? First plate is a droplet of LC. Second plate is a t1.

It's ape.

What recipe are those on? They have an interesting look to em, maybe it's the way the food coloring shows through.

I wouldn't take the lc test plate to grain being that it's right on the edge of the plate.

The other one I would transfer from anywhere and then take the rest to grain.

I think those plates look good Pnin.

I also run pda,  just fyi you can decant out the potato flakes for a more clear final product :thumbup:

I tried straining a recent batch through a very tight mesh strainer I use for my kombucha and it seemed like I was only getting water to come through on the other side. Could you explain how you might do that? Thanks!

I dont do anything fancy. I just use a very large mason jar, something like 600-800ml water for making something like 250ml water batch of agar, couple spoons of potato flakes, stir, let it all settle for like 5-10 mins. Pour slowly till ya get to 250ml (for example). Discard leftovers

After lots of lost prints I would never suggest a mini fridge with a freezer. Its either too cold or warm enough that the frost will start melting into everything else. I just got a drink cooler unit so it doesn't get too cold.

SUNKEN MYC Question

Hi everyone

I have this happen from time to time where the myc looks like it is not at surface but somehow within the agar medium and there are no parts of it in the air. Like it's drowning in fluid or sunken into the agar. i don't know how to describe it better.
In this case it popped up after some time and from that point it continued growing normally on the surface with white fluffy myc.
Happens on PDA, MEA, MYA, and grain water agar, doesn't matter what recipe.

Is there any reason for this and is it normal behavior or is there something wrong with my agar or my process?

I thought it might be too much water on the surface from the condensation (i do no-pour glass petris).
Sometimes when i do a transfer i see a drop of water surrounding it and the myc struggles for some time like this looking like it's embedded in the agar until it pops up. Sometimes it doesn't pop up cleanly with a perimeter that is now out on top of agar but just keeps looking wierd like that and pops up here and there.

Are most of yous using a tiny amount of nutritional yeast in your agar? I only use lme and I never get the super rhizo look. Literally all my plates regardless of variety look like this.

I like yeast in the mix for cubes. I use brewers yeast but it doesn't matter.

Try lowering your nutrient if it is over 1.5% for more rhizo growth.

Ive almost always stuck to 1% but increasing to 1.5-2% made absolutely no difference in growth. Just made more carmelized speckles in the plates.
