Transfer from where you want doesnt really matter those all look pretty good. I wouldnt inoculate grain with just a josex poke let alone a whole bag, although theoretically it is possible

Ok thanks. Should I make sslc then? Do I need to transfer again or can I chance it if I josex poke to sslc directly? I mean I could do both.

Also should I go for the thicker parts on the semperviva? It’s all fluffy, but some parts are thicker as seen by the backlight.

On the nats, might be worth trying to select away from the rizo growth.

I'm not up to date, but I think i recall the rizo growth showing more overlaying nat myc and better luck with the more tom. Mycelium

I would take from the fastest most aggressive colony.

Also, I streak my plates like a few others around here, or do the good ol’ scrape of a spore print with a sterile blade and good ol tappy tap above the plate and in front of my flow hood.

Generally I get spores only and takes one or two transfers to get solidly, clean and healthy mycelium like these.

The plate with a tiny dried up mushroom is t1 away from the MS plate. I let the MS plate die, probably shouldn’t have but… oh well.

The plate with clean healthy mycelium is t2

I tried to clone the in vitro mushroom but who knows. I got a pretty good selection it seems. Super aggressive and quick to colonize the plate. Excited to see it on grain here soon.

Thanks, but I’m torn between the semperviva plates because no one is noticeably more aggressive. They were all transferred the same day from two ms plates (a and b)
#2 is slightly bigger, but not as rhizo as parts of the other two. Hence my indecision. I’m under the impression rhizo is preferable but does that depend on the strain, in this case semperviva?

Are you using josex poke to make transfers? Im confused by your response. Im just saying its bad practice to inoculate grain with a ms plate.

I’ve never had “rhizo” growth from sempervivas. Just take anything that looks healthy and clean.

I’m using a needle as a transfer tool, according to the josex method (although I forgot to fill it with water, so it was literally a poke, no water involved. Seemed to work well but I did do two or three pokes per new plate just in case, which is why you see 2-3 on the semperviva T1 plates.

Ok, so hypothetically I can nocc grain with the T1 semperviva, but transfer the ms natalensis plate.

Sslc can be difficult to use with a syringe. I like using SSLC for free pouring but for this situation I think normal LC is the way to go

One of my plates is at 2 and a half days post inoculation and I see something which I can't show you because the camera won't capture it. But I see little wavy lines of whiteness crossing a small area under the surface of the agar.

Happening? :trolldance:
