Here's one more.

An isolated strain is a paired set of individual monos.

Not to say making transfers isn't "isolating" in a way, but an isolated strain is a pair of monos.

Personally I don't like monokaryon work. I think it is an amateur and idiotic way of working with life. Weak cultures that paired out of extreme desperation and not choice. A task for OCD people to feel superior. You know Tom Hanks was bangin' Wilson from time to time.

I prefer to keep my life forms happy. Isolation is depressing.

1/2 1/3 2/3 if all three monos were compatible. Don't even get me started on gene cluster transfers or how hyphae will "harvest" patterns from others be they live, dead, or still spores.

Without sequencing it is really hard to tell what is actually happening.

Are we just arguing semantics? Because while I understand what the different methods of isolation are, it makes me wonder how we are defining them. How do we differentiate between types of isolates?

not to keep harping on the subject but it's been on my mind after reading the last few pages

so a clone is generally regarded to have more homogenized/a smaller set of genetics/strains right? so why does taking a tissue sample out of a clone achieve this vs taking an agar transfer of a similar size? is it that the hyphae are more swollen in a fruit, therefore making that transfer effectively less strains than you would get from an agar transfer of a culture?

I like the way you think. feel free to dm me if you want to just banter. I don't deal in absolutes if I can avoid it. Absolutism is narrow. Semantics are moot and only tools for understanding, not weapons to wield in a superiority gambit. The ultimate goal is understanding each other to streamline information sharing, thus easing the learning curve and contributing to advancement on a community level. We can stroll as equals if you want friend. Understanding is a literal term. You can't do it if you look down at people and the world (the proverbial "you" not you you).

All this terminology is very, very, very, new. We are collectively creating it. It will change as every vernacular will. That is adaptation.
