nah that's grain prep on point. I have jars that took 4 days to get that level. not too wet, not too dry.:letsgo:

even if its bacterial, who gives a fuck the myc is happy now

Sweet, now to replicate my prep procedure regularly going forward.  I simmered the WBS for @ a half an hour, dried it for an hour and PC'd it.

To test my cultures if I'm not sure I make up small jelly jars of grain, they colonize quick and fit in a small container.

Hey

I'm trying to clean a spore print I did some month ago when I did not know what still air and sterile technique was. I inoculated 10 agar dishes and its a bacteria feast.

I'm trying to find some salvageable stuff. Could that be some weak mycelium ?

I also hypothesize genetics and conditions; it’s a t2 isolation.
If I hadn’t had so many cups look like this I would say it were totally dependent on genetics but as these are clean transfers and I have had so many plates look like this recently, I’m wondering if my malt is aging and just need to replace it. A buddy of mine suggested PDA, which has been useful to me in the past; think I’ll give that a go.



I purchased BULK Malt Extract, so much infact that the one purchase has lasted me about five years.
It could be because the Malt Extract is aging, though it has shown no change in color, smell, or consistency.
I get this growth from other methods of agar too, PDA, LMEA, and simple grain water+agar gives the same results.
I used to just toss pure grain water into 10g agar and it was perfect, but now for some reason it's not working as well for me.




I've picked up some fresh Malt Extract, Fresh potatoes as well, and I'll be making grain water throughout tonight.
we'll see if I can stomp out that wet looking tepid growth.
I'll be making 20 plates of each recipe and recording my findings per plate very carefully.
Most likely I will put up a large post on my procedure and occurrences during this.

I wonder ig you guys can help me here please?

I took some grain from a colonized grain jar for agar before I put it to bulk (to avoid having to go from spores again).

These are the plates form the grain.

What do you think? How are they looking? Anything for grain, or particular areas to transplant to a new plate?

The 2 right side with weak growth look funky. The two on the left look good but not completely uniform growth.

If it were me they would all get transfers

The only one thats really concerning to me is the bottom right plate.  If the transfers you took from the 2 plates on the left (hoping thats transfers and not blank spots on the plate) grow out cleanly they should be ready for grain.

The plates on the left haven't had any transfers from them so far. So as you see there are a couple of patches of dodgy growth. No idea why?!

But I presumably just need to take transfers from the outer edge of one of the healthy looking hits of thick rhizomorphic growth away from the patches?

Do you think it will be ok for me to store these in the fridge for a week before doing transfers? I need to get some plates ready and am away this weekend

That patchyness is indeed odd then.  If you look at the plate from the backside, do you see any bacterial patches that correspond to the lighter growth on top?  Either way take transfer(s) from the leading edge of growth to new plates.  I would expect that next generation of plates to be usable on grains barring satellite contams on the plates.

They'll be fine in the fridge for a week - if you put them inside another container it will help control condensation from the temp swings better.

Going back to PDA in search of the really fingerlicious leading edge stuff because my growth has been wispy and nonchalant lately with my current LME.

500ml h2o
9g Fitlane agar-agar
8.5g dehydrated potato flakes
1 drop Karo light corn syrup
1 drop blue food coloring



pouring agar lazy-susan style on a monday evening feels right, can't wait to fill these up with ghost clones!

Nice setup there.  Have fun, I hope this recipe work for you. Have you tried adding nutritional yeast or peptide to your agar? I have peptide in my agar and the mycelium seems to like it.

Alright so stop me if I fucked up but:
150g potato(fresh, wet, and boiled just like my coffee in the morning)
10g agar
1g AC
1g Nutritional Yeast.

PC for 25min at 15~16psi
Water was 7.1pH when tested before PC and ingredient addition.

Great will do, this is the first of about six different recipes I'll be recording, I have more pics and data but that will be released in one larger post from my journal after it's all been compiled.
Hopefully one of these can solve my bloody problem because I've never had such a bollocks of a time with agar.

What do you think the issue is? What are you doing differently? Are your problems all from the same batch of agar? Different malt, agar powder, nutrients, prep method? Different incubation parameters, temp, gas exchange light exposure, using different material to wrap the plates, phase of the moon, I'm sure you could track it down to something silly that's causing the issue .

Alright so here's the run down.
I've made PDA from fresh potatoes, LMEA from five year old Malt extract, and a couple different iterations of grain water.
98% of my plates have come out looking tepid, this is the only example I have because I ran all the other plates on grain to test the culture to see if it would revert to mushroom or mold out which it then produced fruits.

I'm not sure why this continually happens, normally I would just use grain water or LME with agar and be just fine but this has plagued me for the past 200+ plates so I'm mixing it up a little.
