Probably wrong thread but what’s going on here?



Had this cordyceps transfer going and my dried up roll of parafilm cracked and let in what I’m assuming is gym lut. Spores, only spores I had and was working with at that time.

I got it on eBay from a home-brew supply shop. I always thought this looked like a low nute issue. This is also mixed at 2%, maybe I'll try increasing, thanks!

If it's clean, yeah. Some do better than others, but in general they seem much happier on grain. One of the environmental variables is almost certainly the nutrients like Mycolorado said. I also have LME from a brew shop, so we might be having the same issue.

I do get rhizo growth on occasion though so it's not impossible.

I did a no pour agar run the other day, but I also modified the lids. I know it's not really necessary, but I figured seeing as I was using most of them the to clean up MSS that I'd try to eliminate one more contamination vector (by making it so I don't need to open them during inoculation). I've seen others modify thier reusable agar jars (I'm using half-pints), however there's a possibility those posts were a bit dated. They won't fuck anything up, will they?

I don’t mean to be controversial, but I generally only run 1 multispore plate at a time. I’ll do a streak, take 2-4 transfers, then take 1 to T2 and send it. Clone from there.

If results aren’t satisfactory i go back to spore.

Fuck having a million plates I ain’t got the room lol

From another perspective, I often get contams in plates or subsequent jars, so to account for attrition I do more than necessary plates and jars so I end up with an appropriate amount in the end :laugh:

My first clone that remained viable by the time growth started!

Every other clone I’ve taken has been contaminated and the foreign growth has always started and taken over before the clone even had a chance to grow. We’re still really early on here but I am so stoked to see a clone starting growth before any signs of contam are even showing on the plate!

Also took my first T3’s last week!

This clone would be an F1, correct?

How do you normally take clones? Are you sampling internal tissue only?

I’ve only made 3 other attempts. First was an open air abort thrown on a plate. Obviously didn’t have much hope for that. The second I made the mistake of cutting the sample open rather than tearing it open and the third one I’m pretty sure I cut too deep and came out the other side of the fruiting body when I was pulling the sample.

So pretty much just dimwit hardly capable operator error.

Scrape the tissue out vs cutting if you find yourself too heavy handed.



I searched up scalloped myc, understand it's likely a lost cause. But between the old posts & now, out of naive hope, has anyone lucked out in saving a culture like this? I went back to spore multiple times & this was the best one. If I attempted to fruit it, and somehow got a fruit to clone, would the subsequent cultures be fucked as well? Is it worth the plates to keep transferring further? I'm usually one to cut my losses but I really want this one :lol:

You're making it sound as if it's sporulating green.

Scalloping, concentric growths, wispy etc are what most of us get until we really get down with good and clean cultures. As far as I know it's not even yet clear what exactly causes those patterns. It's not great but it's not terrible either.

Most of the TCs and old hands here would tell you they wouldn't trust that but they strive for excellence and they have a lot of experience and cultures to go back to. If that's all you have, send it and see what happens.

I think P9/Stipe and Cap has done some research on the scalloping and he wasn't too happy with the results (meaning he found it to be some kind of parasitic contam). But I can't find anything concrete. Maybe he'll see this and post something.

I'd love to get some more info on this because I have fuckload of plates like that.
