A tiny quantity of mycelium is all that's needed. One main reason that some use more is to speed up colonization. However speed isn't always ideal. If you have several projects on the go for example, getting things done as fast as possible might have you ending up with just too much to handle at one time.

I can't find it now but I think I remember (so this is as ancedotal as it gets) one of the TCs saying that 1 wedge vs an entire petri (jar shaken as to distribute the agar) resulted in only a couple days' difference.

Thanks for the input! Using a smaller amount will definitely stretch out my mycelium and save on agar cups.  I'll give this a try as soon as my transfers are grown out and report the results.

I often inoculate 3-4 quart jars with one condiment cup and I never even use all the agar.

I usually noc up 3-4 quart masters from each petri and they are fully colonized in about 2 weeks.  If I use a tiny wedge it takes a few days longer to get to the first shake.

I'll use two pieces about that size(right near the bottom) to inoculate grain, pint or quart doesn't really matter. Normally I shake around day 3, maybe 4. Pretty much as soon as the myc has jumped off to a few kernels of grain that can each become their own inoculation point after the shake.

Looking for some help with my first ever attempt at cloning to agar! I took clones from few fruits from the first flush of a grow that went from spores on agar and here they are two weeks later:



It looks like plates 5 and 6 are clearly contaminated. Are they others behaving as you might expect? None of them look very rhizomorphic - what's next...

1) Let them grow out a bit and then store in the fridge or go to grain (I would probably want to store in fridge because I just got a crop and I don't use much!)
2) Take sections of these plates to new plates
3) Something else

Yes, I certainly did ... :crazy: in SAB, flame sterilised the exacto blade etc etc... :confused:

All my clones would always look the same. I learned to.put clone tissue to water agar first then transfer to regular agar asap.

Check this shit out. Two T1 from this germ plate. Almost identical looking germination myc.




Here are the transfers:




I've done around 50 transfers so far from these prints (different prints, different varieties), they all look more or less like the one on the right. First time something like the one on the left pops up.

I mean I understand the one on the right could outperform the one on the left once I spawn them but holy shit look at that difference. Is the one on the left responding to some kind of stress, or is it just genetic luck?

Since I took some pics I'm gonna ask some questions:

A) Are these sectors ? :



B) Why does myc grow in the alternating ring formations? I've seen it in many of my own petris and from other people's pics.

That was precisely how I did it! Except in an SAB rather than flow hood.

Can anyone help me work out what happened?

And why would cloning to agar water help?

A nutrient devoid plate gives the myc a chance to outrun the contaminants since there's nothing for the baddies to feed on.

A. Sectoring usually grows a bit faster than rest of the culture and kind of changes its growth pattern in my experience so I wouldn’t say so.

B. Usually alternating ring patterns like that are from temperature swings. How do you heat your grow space?
