Hey guys. I hope this  isn’t seen as spam, but I’ve just done a stupidly detailed write up for my agar process and I was hoping people could take a look at it to help me see where my problems lie. I haven’t had any success yet, and it would be really valuable to get some insight.

It’s big and overly detailed. No worries if you don’t want to read it all.

I read almost all of it. I had a similar
issue recently in my Zero to 50%
thread...kinda

I had weak slow wispy growth and even
contams seemed to struggle. I suspect
it's something in the agar ingredients.

Despite everything being fine maybe you
should try some different brands.

Go for the Living Jin Agar Agar

And the Maltoferm LME
https://a.co/d/bjcWM6D

I don't know if this will fix your problem
it can't hurt to try and isn't expensive.
These two exact products saved my ass and
my cultures.

Done deal. Ingredients are mercifully cheap, so I’m down.

After reading all your technique and
procedures it just makes more sense.
Maybe you got some bad agar maybe the
malt has some chem in it or who knows
what can happen in industrial food
processing. Rare but who knows!

It just doesn't seem like any of your
explained procedures and techniques are
to blame.

At this point i wouldn’t be surprised. I read here about 6 hours a day. I’ve heard stories about bad agar, so it’s certainly possible. Pinky has given me good advice on how to further tighten up my process, but nothing that would explain a total lack of growth. It’s like things actively resist growing on it at all.

I also use Living Jin agar, plus activated charcoal, and use potato flakes, not malt.

Here's my tek, I get nice ropy mycelium almost every time. (But not too ropy.)

Might be other factors, but this works for me:
https://www.shroomery.org/forums/showflat.php/Number/27551422#27551422

I think of many agar recipes having similar results, but someone else was having trouble, then followed my tek and stopped having trouble and also got ropy plates.

When you say potato flake, do you mean like boxed instant potatoes? Like food? Dumb question I’m sure but I want to confirm. That’s an easy buy.

whip up some mashed taters while ur at it the flakes are not that bad. use a bit of milk and some roasted garlic bulbs and you got a meal.

you could boil potatoes as well and use the water but thats a pain in the ass and your stuck with all those potatoes:crankey:

I still have a 4kg bag of flakes I bought in November last year for potato agar, but never opened it, because light malt and yeast agar is the elite recipe "IMO"

If you dump a packet of powdered ranch mix in your instant potatoes you become a culinary god.

Things got weird here. I had a good clean culture from what I could tell. I did a transfer and dropped the rest to grain. The transfer looks like this. Grain is still colonizing nicely. I just shook the grain before I went to put the transfer in the fridge to chill out for a bit, when I saw how it was looking. Any thoughts? It's tidalwave if that makes a difference.
I transferred and sent the bottom one to grain.

So in theory i should be able to slip this guy into a sterile lc jar and it should work same as agar since is going from one sterile enviroment to another right?

Also I just found some grain cook water in my fridge from about 2 weeks ago is it safe to use since it gets sterilized or toss and wait until I prep some more?

@ wild aussie since that mushroom is pretty mature I don't think I would toss that into LC. You could take like a piece of the stem or something but I feel like all the spores in that cap could make it to where it's basically multispore instead of selected genetics. I could be wrong and feel like I'm not explaining it how I want to but imo it's better to go with a piece of agar over that guy into an lc.

I would take a sample of the stem to clone/LC, and make a print from the cap in a sterile container to make sterile spore print/syringes.

Ok thanks both I wasn't sure about the mature cap so if I'm going to put to lc need to grab when very small

Wow, 15 grams of activated charcoal for a 1.6 liter batch of agar? I put 2 grams per 500 ml and that seems excessive. I've been thinking about dialing it back to one gram per 500 ml.

Why so much activated charcoal? Have you tried it with less? I get nice rhizo growth with much, much less charcoal.

Just send a small piece of the stem in, why would you want to throw a whole pin/mushroom into LC? Personally, I’d suck up some LC broth into a syringe, poke through the stem with the needle, and squirt what ever stuck into the jar. I’ve been doing the Josex poke (needle biopsy) on everything, and honestly with clone tissue the biopsy and squirt plates grew faster than the ones I put rice size samples on.
But also, do that sterile spore print! Idk why, but that is high on my list of things to try, invitro fruiting to get sterile spores.
After a biopsy sample and spore print, eat the mushroom to see how the potency is 😝.  though I wonder if there’s enough nutrients in agar to produce much compounds, or does that just effect size?
