Speaking of coloring, how many drops everyone uses? I put 3-4 drops of black and it turned green after pressure cooking

So a question about activated charcoal. Is there a specific kind of charcoal to use or not to use? I wanted to try charcoal in my agar so I used activated charcoal capsules and did it turn out weird. It kind of separated in PC and even some plates ended up looking like donuts with holes in center

So there's that. Also every one of them that I put anything on went weird AF also.

Any ideas? Oh ya every plate did this but did noncharcoal plates at the same time in the SAB and they came out clean?

I would b a lil cautious with the pe cuz the fuzzyness and the outer ring growing but I just sent some pe to grain that looked identical. No issues

Ty for input fellow human. My millet jars keep stalling out and I don’t know if I should take these another transfer out. I PC at 15 for 1.5 and vent for ten but they never seem to go the distance. Granted I guess I don’t know where my contam may be coming from.

Kek I didn’t have gloves near me so I just used the closest thing :laugh:

This looks a little strange. How do we feel about this tissue sample taken 6 days ago? Fastest growth on any plate I’ve ever had, but still next to nothing compared to what I see here. Also the color in the center of that growth changed overnight and now it looks less like mycelium. Thoughts?

Nice growth! It looks like when you transferred the first one, you may have dropped the transfer sample and moved it to where you wanted it on the plate, but it still looks clean. Other than that they look ok to my untrained eyes.

If the color changed it's not good. If it's the color of the agar changing (which it shouldn't with that color plate) and then changing the tint of the mycelium then it would be worth waiting. It doesn't look good to me though. It looks a little fuzzy and gray which is never good on mycelium. You might be able to steal a piece off the tissue away from the contaminate and put that on another plate.

I’m not sure it’s worth it until I can figure out what the problem with my agar process is. Right now I have miraculously managed to get one of the 20 or so plates I’ve started to T2, but it moves so so so slow. I’m trying a different agar recipe, but growth seems to be moving at the same pace. Definitely not at the pace everyone else’s agar does. Contam, no contam - everything takes weeks to grow. The only thing that seems to move fast is bacteria, which I don’t deal with often, thankfully.

There’s a factor here I’m not understanding. The agar recipe is good, we know it is. My temps are fine, light is fine, the pours are good, my sterile procedure isn’t lab quality, but I rarely get contaminated plates. But nothing grows.

I’m starting to feel like I’m complaining now, but really
I’m just perplexed.

That’s exactly what happened lmao. They were some big chunks and wouldn’t get off my scalpel so I had to try wiping some of them off. I also didn’t have any regular gloves so I transferred these in those big thick gloves that you wear for cleaning dishes…that was interesting to do.

I had results similar to this. Cutting back on charcoal and stirring it in in a pot on the stove to fully dissolve it gave great results.
