Simple I like it.

I've been playing with jars that have bulkhead fittings and caps



It's a bulkhead on a jar lid that a syringe just screws on to with a filter, in this case poly, but I'm replacing the poly with an sfd sticker or syringe filter. There is a 1/4 silicone hose ran down around the bottom


I think for the way you do glc and prints to syringes it would work well and in likely gonna rip it off.

I've been using it for li, lc, glc, easy lc

Nice.. I’ve been playing with the idea of using steam to clean SHIP’s after an iso wipe.. just fitting a tube to the weight port on my presto and running it with just water to make a jet of steam at that I can hit the area I’m about to inject through to further clean that spot.. I have to do some temp tests and probably use a fairly short tube but the temp I get right out the vent after 10 minutes is ~202f. If I can maintain close to 180f when it comes out of the tube end, I think it can be benefial.. lots more tests need to be done.  If anything, the sterilized condensation/vapor will wash away anything left from the iso

I suppose that means you don't have a hood. The idea seems sound, I just don't ever inject through anything.

I mean if you are using the oven bag and pcing the ship in the bag it should be as close to sterile as it can get right? Or is it just that you doubt the sterility when going to pull the glc out since it's sat awhile?

I've been trying to come up with a way to inject into my setup for a needle biopsy without cracking the lid. I'm not really having contam problems doing it. But still would like to never open it. That's what the poly was for was to inject through but it constantly got wet. I initially tried to use a top cap to keep the poly sterile. Like this
But none of the epoxies held for very long the PC temps broke them down. Attempts to weld and braze we're not successful either.


I tried using a short thin needle on the syringe for the biopsy and a longer wider needle freshly sterilized and put into the luer lock bulkhead to keep the dirty from touching the clean but it didn't work.
Once I have a clean lc though these containers basically allow me to infinitely expand them keeping it sterile unless I fuck up. 

I like that I'm able to pull up big 60ml syringes, test them on agar and still keep them around capped and ready while I wait no potentially dirty ship.

No hood.  I don’t have a huge problem or anything.. I just want to be as clean as possible and I refuse to open jars without a hood.. I do fine with just using SHIPs but the problem that I run into is because I use glc, bacteria can build up if I continuously knock up a new glc jar when I make qts from the previous glc jar.. eventually, bacteria is gonna make the culture unusable 5 rounds down the line from SHIP pokes (I can/do combat that by drying the glc jars out) but if I can stop that from happening by using steam so there isn’t an eventual bacteria build up, that’d be sweet.

Yeah, it’s because the glc jars sit out for long periods of time.. the jars in the oven bag, I’m not worried about at all.. they are sterilized.

I like the idea of the capped lid.. I think josex uses/has used something similar.  The problem I have is that I already have a ton of culture in these lidded jars and it’d be nice if I didn’t have to make 48 new lids and the real effectiveness of the lids I use now is the amount of control I have with the GE.. I can tape up all the way or leave them I taped by 40% to dry the grains out after colonization.

They’re also nice for printing because the print dries out really fast and I even put the jars in front of the fan to speed the process up.  I’m actually going to be doin g a thread about drying out bacterial cultures to clean them up.

But anyway.. I don’t want to derail the thread.  I appreciate you explaining that all to me :thumbup:

Yeah I'm not sure a being able to sterilize a ship sure would make that process easier though. If I can find a ship laying around I'll steam blast it and kiss it on an agar plate to see if it's relatively sterile.

Yeah I ripped it off of josex 100% I just can't find any kind of epoxy/adhesive in the states that hold the two rings together well. Temp and moisture break they down quick. 
It's a sound theory and the top cap and its keep the poly on the injection lid sterile until removed. It basically allows you to have a sterile ship without a ship. My execution is just lacking. Though I've found I have no real issues just cracking the lid and squirting in a sample. The bulkhead fitting makes drawing up syringe after syringe easy. Though I'd like to add a check valve to draw in less air from the tubing.

Maybe we should have a all things lc and it's assorted bullshit cousins thread? I kinda feel as if we may be shitting up the agar thread

I gotta say, I’m impressed with these condiment cups.


This was the most half assed unsterile work I’ve ever done and so far zero contams on 8 cups. Half I wrapped the lids half I didn’t.



I actually had plates waiting for me when I came home with these so I opened them to use a couple for hot sauce. I left the torn open bag sitting in a cupboard for over a week before pulling a few off the bottom of the stack and pouring them.

:shrug2:  Seems sound to me, expectations exceeded.

Fuck yea AKA looks like you got the ones with the nice lids too! Ive gotten some different ones from target before, they work but the lids are less friendly to work with.

I dont wrap mine and I havent seen any issues, even with plates that are months old

I feel less like a degenerate now for not using petris.

For those of you who use activated charcoal in your agar mixes, what percentage of charcoal do you use? I tried making the black stuff once, but I'm fairly certain I used WAAAY too much charcoal and the growth was weak af.

I’d be lying if I said I measured it. I just grab a spoon out the drawer and a heaping half scoop. I think I estimated it to be 1g per 100ml. I’ve been hearing some complaints of activated charcoal possibly causing issues. I haven’t tried activated myself to verify.

The amazon listing says activated but the manufacturer has confirmed that this charcoal is not activated charcoal and it’s a very fine powder.

I’m surprised more ppl don’t rinse their transfers in sterile water before putting to grain.. just because myc has grown over an area of agar doesn’t mean something can’t land on it afterwards so it’d appear clean only to have dust on it.. maybe ppl do and I just don’t see it or my reasoning is flawed.  Definitely didn’t see any downfall to it.. there was myc coming off the agar bits the next day so it didn’t water log it or anything.  Idk, carry on!

I’ll take my chances moving a wedge from a sealed dish to a fresh jar 3 inches away before rinsing it in water first. Seems more complicated for no benefit.

If something could land on the wedge from the time the dish was open to when it went to the jar, why couldn’t something land on it after it came out of the water?

Isn't rinsing an agar wedge in sterile water just a less efficient liquid inoculant? I mean that with honest curiosity.

Faht you never cease to amaze me with all the weird low tech high efficiency stuff you do. There needs to be another giveaway where someone gets a flowhood and you should win.
