Re: AGAR ENVY! (Anything and All things agar!) [Re: Stipe-n Cap]
    #27368728 -

Got a few questions I'd like to drop here:

1) I want to test my LC (media bottle, regular lid) by sticking an innoc loop into the broth, then streaking on an agar dish.  Cool or FUCKED UP?

2) Is there any danger in handling your petri dishes too much?  Like do I need to wash my hands or be careful about/when handling them by checking on them 69 times a day?

3) If  plate pins, does that mean it's definitley clean?  Can I be sure that it's cool to "go to grain/LC" with a plate pin?

4) Do I need to wrap plates inside the SAB?  I ask because I can never cut my saran cleanly and it causes me a huge headache.  Gonna get a sharp knife ASAP.

5) I get a lot of edge contams - can this be b/c my hands are wet with iso when handling them?  Also, it's so hard to move them without pawing the edges, lifting the lid briefly, etc.  Is there a good way to do this?  LIke they have anti-slide bottoms so it's almost impossible to just slide them and lift from the bottom.

6)Do you just roll up the empty sleeve inside the SAB to use later? It seems like germs would hop in it once opened?

I would either poor a little or use a syringe to squirt a little onto the flame sterilized loop.

2) Is there any danger in handling your petri dishes too much?  Like do I need to wash my hands or be careful about/when handling them by checking on them 69 times a day?

If I pick up my plate, I always sanitize my hands with iso first.

3) If  plate pins, does that mean it's definitley clean?  Can I be sure that it's cool to "go to grain/LC" with a plate pin?

I have never gone straight to grain with a plate pin but I have put a few to LC. It works just fine.

4) Do I need to wrap plates inside the SAB?  I ask because I can never cut my saran cleanly and it causes me a huge headache.  Gonna get a sharp knife ASAP.

I don't use a sab so I cannot comment on this.

Practice makes perfect. Just be conscious of your movements and make sure that nothing that is unsterile hovers over your plates.

If you are referring to the sleeve that the plates came in, I have stored poured plates in them for short period of time on occassion. Never had an issue.

I’ve been using the same sleeves for several months no issues.

A plate pin is no guarantee the plate is clean. I've had plates with obvious contamination pin before and I've had times that pins from a relatively clean looking plate had obviously enmeshed mold or something similar when transferred to a new plate and grown out.

It's only really a concern for me with like germ plates or T1 plates cause after T1 I rarely ever see contamination but just something to keep in mind. Always good to take it to agar first, that way you'll have the culture backed up as well in case it turns out to be a killer.

For the saran rolls I use a bread knife and don't saw through it straight from top to bottom, you can get a cleaner cut if you kind of score around the entire roll and then cut through in a few decisive strokes.

Hogstomper

#4 - is your roll of Saran Wrap cut in 1” rolls? I use parafilm and I don’t use anything to cut it. I just roll it until it laps twice around the dish then pin down a finger where I want the tape to cut and then tighten the tension of the wrap by rolling it forward and pulling at the same time and it snaps. Maybe I’ll make a video or gif of how I do it.

5g lme 5g agar and 250ml water should be a good all purpose mix.

Wait that’s for ten plates. So half that

I've been fiddling with these large pp dishes for master transfers, allows for a decent amount of surface area to take transfers.

These weren't handled well or wrapped so they eventually got some pink goo but still decent size and worth pursuing.
It's just not great for viewing because they don't come with clear lids.

I find this image pretty surprising. This plate was streaked just under four days ago.

I just noticed these "invisible" mycelia today. But it is 3 cm across! It's so flat and transparent that I had a hell of a time getting this photo: it's backlit but also over a black background. I assume it's mold since it grew so fast, but I'm surprised even mold would grow that fast.

I'm hoping someone can help me out. Does this plate look right? It's about 7-10 days old. This is my first attempt at mushroom cultivation but I've been lurking here for a long time now.

Looks like a normal germination plate to me. I would start taking transfer onto a new plate and go from there.

Thanks AKA! Always appreciate it!

Here's another plate. Looks like there may be some chance of myc on the right transfer.

I'm looking at 4 o'clock.
Any help would be appreciated. I suck and think everything is mold.
