Take a transfer from the slant like you would a petri dish, then transfer to a dish.

Same 2 clone cultures (Mexicube) nothing but Tomentose on plates and nothing but ropes in master slant tubes
I'm assuming it's the wood :shrug:

The only time I've seen Rizomorphs on plates is when I clone entire pins. And they quickly turn tomentose after 1 or 2 transfers

My clones are performing fine, so I'm not too worried about it but I am curious

I can never decide if I want tomentose or rhizo when I get these plates with split growth that all looks good.




Prob end up going with that far rhizo tip at 6 like normal.

I figure I'd ask here instead of start my own thread -

What does anyone think about reusing plates? I've got maybe a dozen plates that never germinated but I have a couple clones I need to take and a bunch of transfers to make but I'm out of new plates. They show no signs of contamination at all :shrug:

I know this is far from ideal but to make things worse I'm leaving town in a few days so I definitely won't be able to get any new ones in time.

Edit: just to be clear I would be transferring directly to the ungerminated plates, not emptying, cleaning and re-pouring in the old plate

Question: I'm hunting down the cause of bacterial spawn bags. If three things could likely cause it 1) bad mycelia on agar, 2) bad technique 3) bad sterilization of grain....

And, my agar transfers show no contams, month after month...

Can I use my transfers being free from contams as indicative that my SAB technique is decent... or are bacterial contams sneekier than visible mold contams?

Ya I had 3 swab plates with nadda, and they're fine with something else.

Contams can be really sneaky. Since I started using a hood I’ve realized there’s a good chance I sucked at SAB and was running a lot of cultures that seemed clean but weren’t.

The only sign anything might be wrong was early molding after the first flush. I’m noticing my cultures look slightly different on grain now with the hood. It could be a lot of other things but I’m leaning towards my stuff before being mildly dirty.

Current agar fun: Malabar germination plate pinning on the left, a few jmf invitro agar fruits I transferred, and some T2 GT looking awesome. I love the way this GT myc looks.

I got a red spot on one of my agar plates after using a tissue scraping from the inside of a split mushie. Only did 2 plates to try this. My first time doing this. I put a small live tissue scraping in a few spots on each plate. Any thoughts on this red spot? Looks to have white branching in the middle
