Is there an advantage to using the charcoal in the mix? I searched a little and read some RR notes on the subject about healthier myc and less bacterial contams? Is there anything else going on?

Not that I’ve seen. Just a color contrast without any additional nutrients added to the plate.

I might give it a try. I've read that mycorrhizal fungi love charcoal and seem to develop more prolifically when it's present in the soil. It serves as a habitat/nutrient storage location for them -- I suppose the same would happen in a petri, but I'd be interested to see if all the carbon particles would act like a grain and be another inoculation source?

So I found some premade agar petri dishes with malt extract agar in my surfing of the google box and ordered a few. Am I going to have to pressure cook these when they show up or will that fuck em up? I'm new to agar and these seemed cheap enough so if this is something that I should not use I wont be out much.

You and me both. Ill contact the manufacturer and ask them. I'll also inspect the packaging really well before I do anything with them

I’m in the process of trying to Aquire another but the gentleman that made it shop isn’t heated so will be a few months before that happens. Wanted to use it in a contest/giveaway



Just wanted to add my input to this for anyone wanting a similar tool.
I started using a metal straw only for my clean cultures, inspired by D3

In high school I used to take a small screwdriver, get the metal part red hot and pull it out of the plastic. Then I'd take my honda civic keys and do the same and transfer it into the screwdriver handle. The result is cleaner than you'd expect

I'm sure this could be done with a metal straw as well. just cut it down to a few inches and I'd think you'd have a nearly identical tool

They won't sell them non sterile so will only answer yes to you, definitely no need to pressure cook yourself. The question is whether they sterlised them properly. You are definitely best off doing it yourself if you have a pressure cooker.

Yup! I did almost exactly that with some brass 1/4" tubing and 2 part epoxy puddy and made this lil guy:

Whenever I used to have something like that it was always 100% meshed-in mold, had me hella puzzled for a while the times that happened.
Might be about the most difficult contam to get rid of. Some people have had success with a hot-pour and I had success once poking it and transferring to pucks, the mold and the cube myc separated themselves and I could save the culture.

Yeah if the t3s I took a few days ago do it I’m gonna try poking them next.

Last year pesa kept doing this and I eventually ran it but it took like five weeks to pin.

Total noob here, trying to go from print to agar. I have PC'd the q tip for 20 mins and left it in the jar, the paper towel has ISO on there. Any feedback for improving technique is welcomed!

Just realized that it was a video. I'm not a fan of swabs. You can make an inoculation loop with a piece of wire and an Xacto Knife handle.

When you PC tools, it's better to wrap them in aluminum foil. That way you can carefully unwrap the tool vs fidgeting with a big jar and you Definitely don't want to be reaching over anything that you want to keep sterile. Shirt sleeves are good at dropping contams all over the place and you want to use slow movements inside the box.
