Yeah it's soft. It's the same on every plate which were transfers from various t1's. Hopefully they're ok

Aside from that being a nut worthy picture, it might answer one of my questions: When transferring, is it alright to transfer the original transfer piece to grain? As long as it's clean, of course? I don't see any reason not to I've just always thought I needed to stay away from the previous xfer for whatever reason...?

If that's not the case I'll have PE, APE and GT going to grain in a few days!

Fuck yeah! Not sure why I was over thinking that...I'll be prepping grain tomorrow or the next day. some of these colonies are only the size of a quarter but they're healthy so I'm gonna send 'em

The best looking rhizo I've ever had:

It was on a 1% BRF plate. My best plates always seem to be BRF, too bad they're opaque.

wretched noob alert:

what causes jagged growth like this?  probably a sloppy transfer?  probably also bacteria since I seem to love growing it

Multiple things going on from what I can see. # 1 being the irregular shape of xfers, very jagged and looks like there may be several pieces laying next to eachother causing the multiple growth points to wall up from eachother. That bottom plate looks like theirs 3 pieces all clumped together and if you follow the sector trajectory lines it sort of lines up with the peaks on the wedges. My recommendation take small as possible xfers from spots circled in green.

Here you see some yellow discoloration from some sort contamination most likely. Can see how the myc got really spotty leading out from that zone.

ive not had any luck with my agar so far. my first dished all got contaminated and my second batch does not seem to want to grow anything yet after 2 weeks. i used red dye though so that could be the reason. i have made some blue ones now and i will be trying with those next.
i will keep waiting for now.
i have some BRF jars colonising, so i plan to take a transfer from those and put that on to agar when i am ready to birth them. will that work okay as long as i do it all using proper sterile technique? can i just cut a chunk out and put it on the agar?

Does mycelium like blue food coloring??? Every time I make blue agar it seems to lose all its blue color quickly. Or will it metabolize most colors??

Following advice from here, I've changed my technique whereas I'm going in hot with a metal straw, and then scooping out that bit with a scalpel so hope that helps me improve. Just wondering, how important or beneficial is it to put the wedge back in a new dish with the mycelium facing down / agar facing up? I'm trying to do this, but find it takes extra poking and sliding about as it often lands on its side etc, just hoping this doesn't leave room for extra contam.

Ya I just did that, kept knocking the little squares over till face down. I read where contams are slower crawling out from under than myc.
I did 4 squares per plate. Squares just worked out better than wedges for the realistate.

I have not found a consistent difference in leap off speed going face down vs up personally (in both cases it just fuzzes out at it's surface all over for 24 hours or so then spreads), and it's essentially inconsequential for contams if done right but great for aesthetics. Taking an extra step to flip it is a contam vector, as are any extra steps ever, but especially so if you're having to change tools or keep media exposed longer to do so. Like anything you'll get better and faster if you keep at it though, so you can reduce the size of those vectors. There are tools that can do the circular poke and flip in one motion, which you may want to look into if you like that look. I bought a cork borer on Amazon for $3 and attached a pen spring to it to give it a try, but personally prefer the speed, flexibility and ease of sterilization of a scalpel blade at the expense of some aesthetics.

That's a bad ass APE plate!

How big of a wedge does everyone use when going to grain? I've got some colonies a little bigger than a quarter I want to get onto grain. Just to clarify, I would be doing one colony per quart jar. Does that seem like enough?
