hmmm pretty odd thing for companys to do i mean it weakens the bag making easy open creases like that. now im worried these are all contaminated but they come from a pretty decent labratory supply store. guess ill have to get some going and just test em out lol

I've got the 8qt duo Instant Pot and just checked that I can get at least 3 500 ml media jars in at once.
Thanks AKA. That is going to be sweet!

These are not quite D3 pretty (the gold standard), but they are my pertyist so far. They are T2 from a petri pin clone of a PE that performed well for me.

I am going to see if I can figure out LC from these.

Tissue clone going straight  to LC should probably slant as well.

Just curious..

How many germ plates do you guys typically streak when you get a print/swab from a new variety/species? Also how many clone plate would you typically fill up with biopsy from 1 fruit?

And then following those, how many T1 plates from best growth?

Finally having good success with agar and I have a lot of prints/swabs I wanna get fruited in the most efficient way possible, so I'm curious what your guys' common practice is. I've had success with only 4 germ plates, but I have a feeling that was just good luck...

Depends on the variety. If it’s a swab set and I’ll streak 3-4 and dunk the swab and leave it in the last one. If spore I typically do two plates.

T1’s I do 3 xfers to a plate and depending on what looks good from the streaks on average 3 plates. From there it just depends on how the myc looks. Might do similar on T2

T3 I’ll go one xfer per plate and depending on how myc looks maybe 4-5 plates

The fastest culture I have ever witnessed. Knotting 2 days after full colonization of the agar plate(13 days after transfer)

This was originally a White golden teacher that completed its first flush in 12-13 days time. This is a clone transfer of a day 5 fruit that popped up.

Attempted my first go at Agar a week or so ago, took a tissue sample. Just checked up on it and it looks hideous, god knows whats growing in there! Quite disappointed as I followed what I thought to be sterile technique, the only thing I can think is I didn't sterilize the scalpel properly as I only had a bic lighter that left residue on it.

Is going from tissue to agar generally seen as potentially more problematic than going from a spore print?

Your scalpel was more then likely the culprit, did you wipe down the nasty lighter?

Buy a torch.

Same problems will arise given improper techniques

My friends who use a metalstraw to pre cut agar transfers. I was about to buy some but I was wondering.

You ever have problem with the agar getting stuck inside?:takingnotes:

Yep, I'm not sure if it was cause I let it cool down to much or my agar was too thick. Not sure but pissed me off. I went back to scalpel till I can get one of those pimp ass punches. Would like the straw to work though.

I'll quote myself from D3's tek thread:

Quote:
I had trouble with transfers getting stuck in the cocktail straws mentioned earlier in the thread. So I found some with a larger diameter (0.28", rather than 0.10"), closer to the punch size recommended in the OP. I've done several transfers and have only had it stick once, and none at all since I started cleaning them out with the brush prior to flaming before each session.

I still have only had one puck get stuck out a few hundred with the larger straws. I have also on two occasions had the myc get pushed down into the cut made by the straw. I'm guessing this is due to the edge being somewhat blunt compared to a punch, but it's rare enough that I haven't made any changes.

The straws have a thicker wall thickness coupled with small diameter it really wedges it in there. My custom tool uses brass, if the least bit handy could fashion a handle for a piece of brass tubing like this

uxcell Brass Round Tube, 300mm Length 7.5mm OD 0.2mm Wall Thickness, Seamless Straight Pipe Tubing 3 Pcs https://www.amazon.com/dp/B07Z98SWVZ/ref=cm_sw_r_cp_api_glc_i_cFceGb9DVZWHJ?_encoding=UTF8&psc=1

I keep meaning to make an acrylic mold so I can mass produce some.

I looked on their website and couldn’t find brass tubing but I can’t imagine they don’t have any.
