What's everyones input on getting swabs to germinate?

Streak a few plates and then bury the swab in the last?

Use scissors or tweezers to cut/pull small amounts of the cotton tips off and drop onto multiple plates?

I've got so many great varieties now on swabs and i'm paranoid because only having two swabs of each variety means i've only got a handful of chances to get something viable before i've lost the variety entirely.

Would love to hear from several people on their best practice for using swabs as long as possible.

Mr. Mushie,

On 12/27, I tried my first swabs.  I streaked one plate with one swab of Ban Phang Ka and jammed the tip of the other BPK swab into another plate.

Today on 12/31, I still don't see much happening.

Good luck to you!

The best luck I’ve had with swabs was by tearing small filament off with your sterilized scalpel and putting that into the agar.

No luck with streaks

Any of you guys have an idea what is this transparent growth on the leading edge of mycelium on agar?

And this one:

The red circled area was transparent before it filled in but it's different looking to the opposite side where the mycelium is white and flows.
The red arrows show the transparent growth.

I could see the rhizo structures form on the transparent part before it filled in as white mycelium. What is it?
Note: These plates have slightly lower nutes than the ones that the transfers came from.

I appreciate the input D3, Tom, and Rafiikii.

I wish they weren't so tricky to get germination from, i'm quite stressed about it actually.

Thought about using some beet juice to replace dye and dex. Anyone tried this?

wish I'd seen this before I went and jammed mine in the agar lol good idea. I'll try that with my reserve yeti swab if the others' doesn't take.

juice from fresh beets mixed with water and agar? I'm not sure the nutrient profile of beets but I'm pretty sure they don't have any starch so you might need to add LME. They'd make a really cool and effective dye though, I've always liked their color. My grandma makes a mean beet soup. Might whip up a pot and see if I can't get some beet water first when I boil them

Yeah, so to replace dextrose and give it color. Still use potatoes or brf

There are some threads about it, and some posts in this thread.  It'll grow myc but the color fades quite a bit during the PC cycle so you won't get beet red agar.

Mr. Mushie,

On 12/31, 4 days after inoculation, mycelium appeared on the tip plate.

Today, on 1/1/21, 5 days after inoculation, mycelium appeared on the streaked plate.

The tip germinated quicker and its mycelium is growing more robustly than the streak.

In 1974, streaking naked was the rage for about 4 months.  At the tender age of 14, I streaked around the block near my house, because it was THE THING to do at the time. It was an embarrassing, ungratifying moment for me and a scandalous, repellent event for my unfortunate neighbors.  I never streaked again.

I will never streak another plate with a swab.

Today I also inoculated two plates with two Melmac swabs.

I used a sterilized scalpel to cut off the spore-laden portion of the swabs.  I wasn't able to remove the severed sections with the scalpel, so I used sterilized scissors to cut off the sections and place them on plates.

What a mess!  Fibers everywhere, resulting in multiple inoculation sites. I look forward to cleaning these plates.

All of the swabs I keep receiving come broken upon arrival, so it's not like I can streak plates even if I wanted to.

Going to get some long tweezers so that I can pull the fibers and stick them into the agar in the future.

lol yea that sucks, there hasn't been a single swab with a wood stick that I didn't receive broken. There are medical swabs with the sticks made of other materials that won't break so easily.

You can still use those man. Half the time I break mine in half to use them and jam it into the plate anyway.

I’m pretty surprised none of mine have shown up broken now, never even thought of that.

If not, id imagine your germ plates resemble a rainbow.
