Here’s a plate that I swabbed some APE onto. As you can see I just made two transfers, but I’m not sure that growth is clean... None of the growth looks great to me. Would you transfer any of this growth? Thanks.

Looks pretty suspect, I’d also take as small a xfer as possible from a swab plate unless the growth area looked really nice. As much as I love the xfer tek when there’s visible or suspected contamination smaller the better.

That said I’d be leary of most that growth. Maybe someone more knowledgeable in spotting contams will chime in though.

I don’t think the left or right colonies are myc.

I would’ve prob taken from the same spot and then tried to hotpour it in a few days unless the growth looks better.

Thanks guys. Also swabbed two more plates in hopes of better germination. This is my first attempt at PE/APE and swabs.

how long would you guys toss foil in the oven for to sterilize it? pre cutting some tiny squares for a weird experiment. going to try making spore prints from agar pins that grew on contam'd to shit plates. when i take the clone it brings along its moldy friends with it so i'm going to see if making it a spore print will produce a cleaner start.

Do just take a piece of the colonized jelly from the top?

Aka what looks bad about that growth?  Yes I agree suspect but I wouldn’t say ALL of it isn’t myc... I’ve done transfers from tomentose growth like that with no problems

Edit:  oh I read all the replies again and realized all u guys are iffy about it too

Yeah just any recipe with less than normal amounts of agar which helps with spores.

I'm trying to get into agar and having a hell of a time getting anything that looks healthy.  For my most recent attempt I made up some BRF cakes with a vendor syringe and tried putting the rest to agar.  Cakes are caking along but nothing much is happening on the agar.  (Pastyplates, Bod's 1.5% BRF recipe.)  I know a vendor syringe is a crapshoot, but would softer agar (or different nutrient %) give me a better chance of success?

(Previous attempts were failures due to poor sterile technique, I think, but even in those cases it took forever for anything to grow on the plate.)

Softer agar and higher nutrients are usually recommended for stubborn to germ spores. If you can’t get them to germ on the agar like that I’d recommend you clone a fruit from one of your cakes

How would you all feel about putting this plate to grains?

Thinking just the 3-9 o'clock region.

I'd make a couple transfers from that Rhizo growth and clean it up a bit.

Those look great Ababypheonix!

I have two T-2 plates that are looking nice.

Would you spawn these to grain, LC, or not all?

Ufo revert clone plate pin ripping through the agar at just short of two days.

I would keep cleaning them. Try a hot pour

Do you mean transferring to a newly poured plate while the agar is still hot?
I haven't run across this TEK yet I guess.
