Mine was for sure related to the agar. I had multiple cultures and species react the same way.

If it had only been one or two plates I definitely would’ve thought mold though.

I will cook up some agar again tonight, but isn't that 1.4% nutrients, 7g lme/500ml h20. Maybe I'm not doing that right. Either way I have some ape, maz, and rust spore plate pins I'm going to transfer. I have always used pretty close to the same recipe 7/7 agar/lme to 500ml h20, with pretty good results in the past. Eventually I'll find the golden ticket. Thanks alot everyone.

And yea I have messed with nutes to get different results previously but all the plates look the same relatively to the other transfers, but maybe a little stiffer recipe isn't letting the mycelium penetrate the surface making it look different.

Maybe try distilled water? I find my tap has varying levels of chlorine and some days when I run the shower the chloramines are so strong it’s made my eyes burn just sitting in the bathroom.

I strictly use distilled for my agar as I’ve noticed the few times I used my tap I had poor results.

Have you seen different growth between regular bottled water and distilled?

I usually just use bottled water or filtered tap water and my agar seems to do okay.

Going through the stack of old plates and finding some plate pins. Some  of them will be going to agar tomorrow.
The distilled water seemed to make my agar more clear, I do have really heavy water so maybe that was causing issues, we will see.

That’s the smarter move. I’ve had a couple plate pins I dropped to grain and forgot to put some seeds to a plate when I spawned. Then they were bad ass cultures but I had no way to keep them.

I did not know that about using LC.

Question: If I get pins on plates that I kept after a transfer that show no contam, is it still safe to think of these as sterile? Or does the act of opening the plate for the original transfer disqualify them?

Your transfer would be dirty if the answer was yes.

Not to say you cant contaminate the dish but the act of opening to make a transfer shouldnt dirty it up.

Guys I'm having a hard time cleaning prints lately.
If i'm not sure wether to throw a plate to grain, would it be a good idea to let it grow and pin, and then take the tissue of that sample to another agar? And using that strategy as a way decontaminate the plate (assuming the inner tissue is sterile)? And of course, transfering once more if needed.

I'm going to keep track of where, over the plate,
the media bottle is when pouring agar.

I'm still getting "immaculate edge contamination"!

These edge contams are so consistently alike.

I recently slowed down my hand movements, including lifting the lid.
I pour a stack of five. Try not to slosh the agar.

One thing, the media bottle did suck air just before pouring, but
I still had AL foil and an ISO towel over it when I cracked the lid in the SAB.
I think I  wiped the threads with ISO too.
