Close...it's a strain of PF Redspore I've isolated...drops a reddish purple print instead of the usual baby shit brown that Redspore normally drops.  If you let the spores fly the whole tub turns a burgundy wine color.  Hehe.  Here's a print next to a normal Mars variety print.

I thought it was best practice to let jars get 20-30 percent colonized and then do your first shake. And then that's all that is needed until 100 percent colonization and then shake a second time to break up for spawning... Am I going by outdated info?

Yea if you’re getting those kinds of contaminate rates I’d strongly suggest stepping back and evaluating your clean work method and the space. Is there vents or fans running moving air around. I’d concider showering just before sab work. Are iso wiping gloves, plates , tools ect? Slow cautious movements to not disturb air. I can move the way I do as I’m working in front of a flowhood. Best $400-$500 i’ve spent. Only time I see contams is swabbing plates and that’s from the swabs/prints source.

I shake at 5% if I noc with a little baby wedge, then shake again at 30%. Not sure what the new standard is, but this works pretty well for me.

As with everything in this hobby I started out taking every precaution and have slowly gotten lazy and figured out what I can get away with.

The main thing for me is not bumping into the SAB at all, even a tiny tap seems to cause problems.

At this point I’ve been doing SAB work with no mask or gloves and a shirt absolutely covered in dog hair half the time. Keeping stuff raised off the floor and moving slowly and super careful not to bump or go over anything sterile seems to take care of 99% of my issues.

My most recent idiot moment was a month or so ago when I’d been having a lot of problems and then somebody here pointed out a guy had his phone playing a video on top of the SAB. I’d been putting on podcasts and leaving the phone on top of mine too. :facepalm:


Similar feeling to trying everything to get a new machine to work and then realizing it’s not plugged in.

I love how lazy I’ve been able to be in front of the hood. When in a sab I was iso wiping it down top to bottom inside and out, everything that went in got wiped down, fresh clean towel underneath ect ect. Shower before doing my work, wipe my arms down, a lot of breath holding. Now I just come down to the lab, grab a paper towel, drop my scalpel and punch on it. Iso them, drop into my glass in front of hood so they can dry and use the iso soaked paper towel to wipe my work surface down and my worklight hanging over flow just for good measure. Whole process takes less than 2 mins and I’m off and running. Only time I wipe my plates down is if I’m transferring to grain as I am bringing the dish over my jar and flicking wedges out. Figured better safe than sorry.

Something else,  breath heavy like that fat bastard I’m becoming and easily get away with it.

Here’s the filter and blower I’m running. It’s the wood that hurts the pocket book.

I turn my phone on airplane mode and keep the flashlight on and it sitting on the top of the sab playing music or whatever.

next time somebody looks at me funny for buying a lab grade flow hood before I'd ever germinated a spore, I'll link this post.

Also, gives me hope that last week's agar/spore efforts will be successful.

If you’re planning on running the same culture a lot then lc is perfect. It expands so much quicker than myc on plates and stores better too.

I built my hood before I ever had a harvest. Didn’t take much at all to hate using a SAB

I had plenty of success with my sab... but when I decided to move up to bags a hood was a no-brainer... and I couldn't be happier... mine was only about 400 cuz my dad had an old furnace blower that was perfect for my needs... i love it!

This is why you don’t play in the lab when you been drinking tequila all day. Right after the video I tripped over my rocking chair leg. When I caught myself I dislodged all the tat transfer pucks having them roll around in their petris 🤣😩. Don’t mind my sloppy work. Wanted to show you how I do my prep but turned into a video of what not to do lmao



PS I hate these new celltreat petris. Paper thin and crack if you look at them wrong. Previously I bought a 300 pack with the grip ring and they was awesome. Thick, durable, stacked nice screamed quality. Only down side is they where in packs of ten which was a pita. Thinking the 500 pack with sleeves of 20 would be equally nice I went ahead and ordered some. Sorely disappointed. Not sure why such a quality difference.

Don't get too caught up in it like I did. I spent weeks making transfers trying to get that perfect plate when I really could have dropped a wedge and been over the hump. For whatever reason, I was under the impression that I needed a full plate. Plates kept contaminating before full growth and just kept transferring. Then this happened.

The trick is not waiting too long to start shaking. To give you an idea... if the piece of agar is the size of a dime... when the growth looks like the size of a quarter i'm shaking if i havent done so already. My first shake is usually maybe 3 days after knocking up the jar.

Once myc grabs onto a nice lil cluster of grains around the agar... shake. Then like 3 days later... shake it again. Your jars should finish faster.



Utilizing that advice, I dropped some wedges about nickel-quarter size and waited until they looked like this.



I gave 'em a good shake and rolled the jars around to get the colonized oats to distribute a little better and when the myc grabbed onto another cluster of oats, they got shook and rolled again.



Once the colonized oats looked pretty well distributed through out the jar,

I left them alone for a week and came back to this.



Some of the wedges that went into these jars came from plates that had mold and bacteria growing in them. Obviously you want to stay well away from that part of the plate as to not accidentally touch it with the blade and work real slow and careful or your jar will end up like this.


Don't forget, if you have 1 good jar, you potentially have many more of the same culture if you G2G.




This is a nice TEK for sure. Those jars look great!
