I have always went pretty straight forward with plates, equal parts agar and lme

looking to promote greater rhizos, so lets say I usually do 10g agar - 10 g lme

what's an ideal lower amount of nutrients to promote rhizomorphs?

I've been doing 9g Agar (makes it a little easier to move the transfer imo) and 8g LME. The rhizo is there but I wouldn't say its enough to promote rhizomorphic growth. Try 6 or 7 g LME. 🤷‍♂️

Agreed. I wasnt quite thinking half lme, 7 sounds about right.

What is this pink/purple contam i keep getting. I tried to incoulate agar with a syringe for the first time and everytime i get quick growth that turns pinkish purple but otherwise continues to look like myc. After leaving for ages it looks dusty and pink. Its visible in some of the direct inoculated grain jars too i assume all as all the agar got it.

I'm pretty sure the syringe is actually ps. Mexicana so i don't expect much rhizo but the tometose looking growth has characteristics of cobweb mould also when it initially grows white its wispy and grows upwards.

I cant find anything like it in contams exept perhaps red bread mould

Edit: pics of the 3 grain jars with the same appearance in some spots

To get rhizo growth I had the most success with around 5g lme to 450ml, and then pouring the plates thin.

Anyone have any tips for picking up entire plates of agar with a scalpel? Particularly if it's a softer agar recipe like 1.5%?

I'm working with 100mm dishes, going to blenderless LI. The first time I did it I only used half a plate and managed to pick it up alright but it was a 2% recipe. I'd like to make a bunch of LI jars with one plate per jar and want to be able to transfer the entire plate of agar instead of doing two half plate transfers into the jar to minimize the amount of times I'm opening the plate and the jar.

Seems like you answered your own question... just make your agar a little stiffer, you'll still get plenty of noc points with the mycelium that breaks off into the water...

My go to recipe is 20g agar, 13-14g lme and 1000ml distilled so roughly 6.5g -7g lme for a 20 plate pour. I really want to try my hand at LC and might pour a small batch of ultra soft plates so I can do blenderless.

Well even with the stiffer agar I'm not confident I'd be able to pick up the whole plate, and regardless I want to use softer agar. I'm aware stiffer agar creates enough inoculation points when shaken but the last time I did it colonization was a bit uneven between my jars and I'd like to break it up to get as even of a dispersal of myc throughout the LI.

Not the end of the world if I have to drop two half plates into the sterile water but was just looking for tips people might've had about it.

using pastyplates for the softer agar is good tip. Allowing you to tiger drop it right into the jar.

Yea, this. Or if have to use petris I trim around outer edge with my scalpel, and tiger drop whole puck with a gentle flick from the blade. How I knock up my jars of grain if I don’t pizza quarter wedges

True, I'm just relatively new to agar and I started with pasty plates and I hated the lack of the visibility. My first experience with agar, once I had started using actual petris, I'm almost certain I ended up with like 95% contam rate from a very sneaky mold piggybacking on all my plates that I couldn't transfer out and I'm not sure I would've caught it at all if I was using pastys.

Regardless, I'm a bit more confident now though so I might just clean up the culture on regular petris then when I'm confident it's clean take it to a very soft pasty plate as a final transfer before LI.

And @Demonic, cutting out the outer edges seems like a good idea, gonna fuck around with some blank plates and scalpel to see how if I can confidently pick it up that way.

Ohhhh shit I totally wasn't getting what you were saying at first but that's pretty ingenious. Solves the issue of picking it up and I can cut a circle of agar smaller than my jar mouth. Thanks dude!

Thats exactly what I do. Clean culture up on regular petris and expand to pasty plates to Tiger drop 2.0 them

Before I even open up my pasty plates I loosen the agar from the plate by pinching it.

Then it falls right out like g2g.
