Re: AGAR ENVY! (Anything and All things agar!) [Re: Smoothcat]

I forgot about my no pour plates in the PC and took them out to cool a little late and caused tiny bubbles in the agar. My question is can I resterilize them to get rid of the bubbles?

Have I achieved isolation? [Re: RatsboggleBiologic]
    #26867214 -

Looking for opinions on whether this is an isolate, or if I should transfer additional sections. This is my first attempt at agar.

I dug out some old agar plates the other day and one had these little gateways too. Believe it was a 2nd transfer.

Thats not an "isolate" but it looks like a good non competitive colony.

Those look like primordia which can turn into pins. To continue growth and not risk them dying and rotting, I'd take from a section of agar where those aren't.

I really dig this picture, is there a way I can copy it to my Gallery?

I stopped using my bacti-cinerator after buying 10 scalpel handles. . .
Set up 10 blades on handles. . . Wrap each in a foil pouch. . . PC standing in a jar. . .
Now to whole handle is sterile. . . Cheap aliexpress handles are fine. . .
They can be used months after being sterilised if stored proper. . .

If I can't stop infecting fresh plates, I might try that but...
What kind of handles are you talking about? I'm using X-Actos (2).
I'm suspicious of the knurling on the handle being hard to keep free of contams.

I use #7 scalpel handles. . . Nice and long. . . $1.50 on aliexpress. . . You can flame (or bacti-cinerator) sterilise as much as you want without handles heating, though I like 10 sterile scalpels just sitting waiting. . .

Seriously nice looking culture. Looks like a snowflake. I just had to throw out my golden halo print after wasting at least 30 plates trying to germinate anything. Tried different things, just wasn't meant to be I guess:shrug:.

First plate just looks uber tomentose and disorganized... fairly normal for a tissue plate IMO/E

Id take it a few transfers out personally

Clone plates of this monster.
Hopefully what caused those to look odd is genetic.
If not then I've got some beautiful myc anyhow :stoned:

Those are cool looking.

What’s your cloning method? Removing the rectangle from the stipe and then taking a tiny piece from the hole?

I’ve never seen anybody do it where they don’t crack the stem and peel it in half.

Normally yeah I'd take it apart and select cuts from within the stem
However I wanted to preserve it cosmetically if in the case I can't replicate them.
I just chopped off the outer layer, fillet it and then bored a cone-shape which came out easily.

Efforts to get a clean culture from a swab of APE. T3 here. Still very dense Myc, is it normal to have the faint circle of what may be reach Myc around the dense?

I’ve noticed ever since I started using LME I don’t get nice rizo growth like I used to with the same amount of BRF. I may switch back to BRF because I’m on my second transfer and am mostly getting tom growth. Anyone here like BRF over other nutes?
