All of those are looking excellent!  If it were me, I'd take a few more transfers from that last plate and drop it to some grain! The transfers are just to keep that culture goin on agar. The first 4 I'd let em get about a 1/4" from the edge and then drop em on grain also... good looking plates dude!

Thanks man! I really appreciate it.
It's a relief to know that they are fine. I'll go and cook some grain then

The mycelium came from a plate, I took a needle biopsy from the leading edge of a maz t1 plate. But it was fast and it will get LCed eventually once I think its clear.

Crazy. . . How many days was that from biopsy?

Hey ya'll, what do you do with germ plates after you make transfers?

If I find a culture I really like I will slant it and toss the rest of the plate in my outdoor patch.

Depends on the plate and it’s reusability, but typically scrape them out and wash them for the next batch. Most petri can be reused, but it’s not fun cleaning them if they can’t be PC’d. PP5s,  just make a big bleach bath for them to soak in and then if they had something really nasty in them, chuck them in the pc for 30min to make sure they’re disinfected.

Might not be standard procedure, but it’s worked fine so far.

This too of course :super:

It's a pp5 and looks pretty clean. This was taken yesterday and it's still nice and white. Although I'm struggling to get good clear photos.

I made 3 T1 plates which are colonizing the transfer chunk at the moment. White and fuzzy.

Typically you want to take that slant sample at the same time, so you aren't returning to the scene of the crime. Even though it looks clean, you'd be hard pressed to know if any bacteria got in there.

That said, you could transfer to a slant and see how well it does and if it fully colonizes the agar and the wood, then you should be fine to store it long term, then just get rid of the rest or recycle it into an outdoor space like Sockadin said

Ok. I think I'll probably just toss it in my compost since I don't have a vessel to make a slant. I've got plenty more of the spores anyway. Another question, I know most people go all the way to T3 or beyond before spawning to grain but is a T2 safe if it looks like healthy growth?

The more you cut it down, the more selective you're being about the genes you want to use, but you could try and get away with it. If it's grown out to about a 1/4 " from the edge and everything looks healthy you could certainly try take from the running edge. The more transfers, the more likely you have a clean and functional rhizomorphic specimen and that much closer to a true isolate.

My buddy, Krombopulos, highly recommends getting some slant stuffs for your long term storage needs.

Screw-tops were $35CAD for 50 and the dowels were $7 and they cut into perfectly fitting halves that fit in the container on a wedged angle, so the myc can reach off the agar for it.

I'll look into getting some when the lab supply stores open back up again. Typically I don't/won't order from Amazon because A.) it's a shitty evil company and B.) shipping to South America is ludicrously expensive, even before the high import taxes get applied. That said there are plenty of chemistry stores in the area although they're closed down for the quarantine.

Oh man, I can dig that. As much as Krombopulos hates Amazon, he can't hate on their prices and shipping during our stay at home and its wild how much of this stuff they have available.
Don't trust them, so everything is sent to a buddy's place and then he picks it up from him once a week and he uses a router built VPN while ordering, so he's not worried. Not doing anything illicit anyways, as his research is purely based on a biological and neurophysiological interest anyways :naughty:

Check out this fluffball on my P. tampanensis galindoi plate! It's thin, fluffy, but still looks like it could be Psilocybe myc. Thoughts? The other myc on these plates is denser but still tomentose.

It's growing tall/spherically:

Right at the edge of the plate. Contam?

Looks like dandelion fluff

Yup, looks just like fluffy myc. Let it go a while and see if you can get a clean pull from the edge closest to the center to transfer out.

Would attempt to transfer some off those two smaller solid spots too as they look a little more gung-ho.

Damn, man. Try looking in craft stores too, a lot of the time they won't look like this, but you might find little storage tubes they say are for beads and shit there. Just try make sure they're PP5s.
