I tossed 4 jars and kept two out of curiosity. They’re colonizing well. I’m not sure that it was bacterial. Frustrated I tossed the jars I did. Once fully colonized I’ll put em to a shoebox and see what happens.

Thanks! Sadly most of the actual fruits I've gotten from this particular APE have been less than ideal, but man... they sure look pretty on the plate!

Mix it with a heavy fruiting APE. Like make a jar of each and shoe box. Then clone.

A buddy of mine who is getting back into the hobby is gonna get me a new syringe from a vendor... I've helped him get back into the game so he said he's gonna pay me back... lol. Hopefully I can get some big white wieners from that! Lmao

A maz plate pin I took a biopsy, its crazy how much mycelium gets caught in the tip of the needle. Probably going to do another couple transfers and im going to do a couple LC with it.

I assume this is bad bacterial contam from the syringe I used (former shroomery sponsor found on the vendors page). The other 3 dishes I inoculated with this syringe look the same. I've contacted the vendor to replace. Eek!

None of the other 16 plates (4 varietals x 4 plates each) I did that session show anything similar.

I used the scratch pattern from c10's tek, and it is cool to see it "working", even if it's all contam.

Just a side note, but I like having my plates sit around for some time before using them.
As a noob I'm still getting 1 or 2 plates contam by themselves, so when I take a "mature" plate, I'm confident it won't be the cause of contam.

I'm pretty new here and also new to agar. It's been an amazing journey so far. I started with a pre-colonized growkit that didn't work out => made me frustrated => spent an enourmous amount of time educating myself reading TEKs => decided to dive all the way in => now I have a lot of agar work going on and I can't wait to inoculate some grain. I want to make sure that I have a clean culture first. I'd be grateful for some feedback on my plates and how to best continue my work. To me they look clean, but I've read a lot about hidden contaminations on agar - mold intertwining with cube mycelium and such - and it made me really paranoid. Sorry for the flood of images, I thought the more I'd show, the more chance there would be someone could point something out that otherwise would go unnoticed.

Some more concrete questions:
Which dish looks most promising?
Should I wait for the best dishes to fully colonize and transfer to grain, or are more A2A transfers necessary beforehand?
Could I use the last plate (E) to inoculate grain, or would that not be advised? This plate was already used as a donor, but it looks clean to me.
If these plates were yours, how would you continue the process?

All of those are looking excellent!  If it were me, I'd take a few more transfers from that last plate and drop it to some grain! The transfers are just to keep that culture goin on agar. The first 4 I'd let em get about a 1/4" from the edge and then drop em on grain also... good looking plates dude!

I do the same. These plates sat for three days in incubation after pouring. I'm certain they were clean before I inoculated. Plus, as is evident in the picture, the contam is only where the syringe material was spread with the inoc loop, which was flame sterilized, and it's very heavy contam. I'm 100% certain it's the syringe.

Looks like you may have hit a little bit of gill :shrug:

I did a needle biopsy and reckon i hit some hymenium and got multispore. . .

The mycelium came from a plate, I took a needle biopsy from the leading edge of a maz t1 plate. But it was fast and it will get LCed eventually once I think its clear.

Someone say agar?

Finally Krombopulos Micheal has all his ducks in a row and a ton of spores to birth, so now he's off to the races! LC in the fridge is Brazilian and he will be working to clean it up and get it healthy again.

He'll be heavily focused on Australian and GTs, then for kicks he'll be working with B+, Blue Meanie, RustyWhytes and Tasmanian (another favorite).

He has a few different types of PP5s he'll be testing out tomorrow to see which will work best and tons of slant materials to work with. Got that good food coloring so he can play around with MAE and PDA and perhaps some BRF down the road when he needs to switch up. Stoked to try some black plates :grin:

He'll be doing no-pours & EzLCs namely, but some poured plates for comparison of ease, and then clean transfers to LCs and to slants for long-term storage. Ideally he'd like to have 3 slants of each put away for later and a quart of LC for each, that he can keep rolling for the next 6 months.

All the tools he needs finally, from Alc lamp to steel scalpel and few different types of loops. Krombopulos's favorite new embellishment, is his German stainless slant rack. Gotta get that drip drip!  :rolleyes::lol::mushroom2:

(side note, that is 99%, but he has a drum of distilled on hand, and plans on rubber banding the sleeves to his arms in such a way that they won't create turbulence working in the SAB  :thumbup: But Krombopulos has hairy arms and doesn't trust them not to fist his activities in the ass :uhoh:)
