Hell yeah man especially from spores. I’m definitely grabbing a piece from 3 or 9 when I drop it.

I was so happy it cleaned up quick, some other ones I’m doing now like b+ are on t3 and only halfway done.

The power of tiny wedges!

Did they pin fast for you??

Haha yes. I’d like to think it’s my sweet swabbing skills but probably luck more than anything

They always pin around 12-14 days for me.

Usually a one flush wonder tho. Occasionally I’ll pull a second or third flush but not often.

Guys,

I'm in the process of attempting to isolate a Stropharia cubensis strain.  I'm on my third transfer.  After the transfers, I simply threw away the first two pucks.  That felt like a senseless waste.

I don't want to devote a quart jar of grain or a tub of bulk substrate to them.  I was considering putting the puck in a half pint jar of CVG to see what develops.  Do you think they would grow and fruit in CVG?

What do you do with the pucks from your first few transfers?

Thanks,

Tom

I usually keep them Incase something happens to the newer ones, or if I definitely don’t need them anymore use them for experimental stuff.

A lot of guys use brf cakes to test clones.

You could mix coir and brf then mix the plate into it, or make li from it.

I’d probably make a half pint of grain and do a small tub or coffee mug/bottle type grow to test it in your case.

But you’re way off from a real isolate if that’s what you’re going for. They can take like 100 transfers and then suck anyway.

I’d put i to a deli cup with a little grain and once the grain colonized I’d mix in some sub and fruit in the deli cup.

Thanks for the fine ideas guys.

But I'm still curious about putting CVG and the puck in a half pint jar.  Wouldn't the mycelium colonize the CVG in the jar just like it colonizes CVG in a bulk monotub?

If it wouldn't colonize, can you explain why not?

Thanks again.

Yeah it will, but the issue is will the agar have enough nutrients for it.

Idk it might, but that’s why I suggested maybe adding some brf or something.

Ah yes, the nutrients.  That makes sense.

Thanks.

My little collection

New to all this so this lot is from spore, currently on 3rd tranfer, also just took a few clone samples in there.

Tried Petri Dishes, small jam jars, and 2oz ketchup pots.

Petri dishes are great if you have parafilm, I don't so I used micropore tape, PITA to take the tape off with gloves in the SAB since it shreds the gloves. Jars are great for no pour since they can go in the PC, but a PITA to get a sample out of, and not very easy to see whats going on inside. Ketchup pots have been easiest to use, once you realise you have to stack them upside down or the nipple in the middle of the lid will collect all the condensation and drip it right on top of your sample.

Also tried food die but the stuff I used is probably organic since in the PC it disapears, when I added it in the SAB I got contam, mycelium also seems to eat it as the colour disapears as the culture grows. Looking for another source/diff brand.

That's the sum total of my experience

I slice however big I want.
To explain I had other plates that showed contamination so I took the section of myc furthest from it.
Saved as much myc as possible, ya know because mercy.
Theres one there where I Just chopped out the segment of agar that had contam and then even took a huge cut. Was fun.

You can use cling wrap instead of parafilm. Cut a piece wide enough (petri+10mm) off the roll with a super sharp blade. Then you have a mini roll just the right width 👍

I used to use food colouring. . . Now just one less thing to have to worry about

I'm gonna compare some huge pie slices to scraping with a needle in a few days.

I have actually done this side by side with these plates.
One plate I took the two huge transfers off of
Another I took a small pinhead transfer by just scraping off the myc.
Between the two the pinhead is much smoother rounded growth and moving quickly.
While the larger cut is taking a bit to recoup.
The pinhead plate is in the lower right hand side of my pic, you can easily see its origin point is a little pinprick of myc.

Testing out of curiosity.
8+pH with fulvic trace minerals
Bottle is 500ml + 5g LME + 9g Agar Agar
Rice hot pack works pretty good to mitigate condensation.
