incubate agar plates

ey! since i been messing around with agar recently (and it all works REAL good) i have been wondering if one could actually speed up the extremely slow colonisation rate of peroxide agar by incubating them. i know, that on peroxide agar the growth usually is slower, but since i store mine at quite chilly room temps they are growing at ridiculously slow speed. now, i was wondering if i could simply put them into the incubator where there's a nice 27.5C/81.5F all the time to somewhat speed up the slow colonisation. i understand that myc really likes this temp for growing in general, so it should also like it on agar right? hope for input peace ohm *edit* i forgot to add that i hit the search quite a bit and didnt find the answer to my question. i think this should also be covered in the AGAR section of the mushroom cultivation archive.

allright since there was no reply i spent the last hour searching and finally found a reference to my question.

It is now necessary to let the spores incubate. Place the agar filled petri dishes that have been inoculated in a dark, warm (86 F) place for a week. After a week check on them, they should show signs of growth. Over the next week or two you will start to see white mycelium forming and growing out from the center of the dish. During this phase you will probably notice that there are different types of mycelium growing. Some may seem strandy (rhizomorphic) and some may seem cottony (tomentose). The characteristic of different types of mycelium growing on the same dish is called Sectoring, this is normal and expected. Keep an eye on the dishes and look for mycelium that looks especially fast growing and that is very rhizomorphic (strandy, rope like), this is the mycelium that we want. You do not want to let the mycelium reach the sides of the dishes, as the sides of the dishes are prime spots for contaminants to be lurking. Once the dish is 3/4 colonized it is time to go onto the next step.

ARGH ! after digging the mushroom cultivator, i came across something else!
on page 37 he goes about incubating the slants at 24C, but i guess incubating them around 27.5 cannot be that bad can it? just wanna do the best to my little cultures, not wanting to grill any of my babies  INPUT !!! peace ohm

Every higher temperatures will cause a profound sweating of the agar. I keep my agar at room temperature, I am not that in a hurry

heh thx for the input. if one is just about to store the petri in the inc till its about 90% would that also be a mayor facor to water content loss or should that be ok? like remove it after that point and store at roomtemp. peace ohm

llright i decided to simply store them at roomtemp one remains in the incubator for testing. should i turn them upside down? i remember reading that somewhere. peace ohm

yes the people usually turn them upside down,it's good if there is concedation. i did yesterday my first agar work. i put most of my petri dishes in my incubator upside down

You can get petri dishes with vents that help with condensation on the agar.

well no i didnt use H202,i though that it's my first time,i shouldn't do it complicated!! Even though as i know h2o2 is needed when u r gonne inoculate with a fruitbody, i did only with multispore solution. How much h202 did u put in each petri? and i guess u did it after sterilization(or else u have killed the h2o2)

i measured out 100ml prior to sterilisation in a filtered small jar, let it cool till 'holdable' then injected 1ml 30% h2o2, stirred and poured. peace ohm

ok i will check now your project one question: when u say 1ml 30% H2O2.... you mean 1ml from a 30%h2o2/70% water mix?
my own question seems foolish to me! lol. maybe i drunk too much for tonight

As long as you don't go over 90deg it should work fine. :mushroom
