What are the pros and cons for high vs low nutes and soft vs firm agar? Is it true it's better to use soft agar when inoculating with swabs? Lastly, what do you look for in a plate before it's ready for a2g? Uniformity/no sectoring? Rhizo growth? Or as long as it is clean?

Depends. If you wanna get something to grain asap, any clean plate (maybe except germ plates) will do. Lately people have been experiencing that tomentose myc can be just as good of a fruiter as rhizo myc.
If you want to have an isolate or something close to it before you put it to grain, wait until your growth shows no sectoring.

I've even dropped germ plates that were clean... lol. Also, seems a lot of peeps that are new to agar all want an isolate or a monoculture... those things are great if you pulled clones and did xfers and all that yourself... you're not gonna get an isolate overnight...

I'm a little worried. My T2's are being super slow. 6 days and they've hardly jumped off the transfer wedge and they look quite whispy and light. Should I be concerned?

I tossed all the T1's too like an idiot. I still have the germ plate though and it's looking fine. I took a provisional transfer from it earlier today so I might not be too far behind the eightball.

I've made a transfer from this petri:


to this one:


As you can see there are two spot of something growing,
this is the piece of agar that I've transferred:

I see the mycelium too fluffy, maybe is cobweb since is there another spot?


I'm near the 'agar to jars moment', but idk what to do with these dishes

yer gonna get schooled for using imgur...I could be wrong...

When you upload pics to your Pics area, personal data is stripped from the pic.
And you can create Folders to keep yer pics organized.

That second pic could be mold.

Anytime I see a little spot with a bright white dot in the middle it’s been mold. Especially since it’s near the edge. Keep an eye on it and do something before it sporulates.

^^^also that. People won’t click shit off site.

Ive read that some people try to have a single spore culture which I suppose they want to link up with another single spore.

Can you see a difference in the growth or mycelium characteristics between monocaryon and dicaryon or is it plausable to assume that if you used a syringe or spore swab that the mycelium growing is dicayotic?

Mind you, I am reading here that tomentose can also produce fruit and not just rhizomorphic?
So maybe im worrying about things that is really insignificant.
It just gets frustrating as fuck when you're not entirely sure what to expect,follow a description best you can, tick the boxes, get pretty damn close

Tormentose vs rhizomorphic often relates to the amount of nutrient in your agar. . . High nute often makes tormentose as the myc isn't in a hurry as it has plenty of food. . .
Rhizomorphic happens when there isn't much nutrient and the myc is on the hunt for food. . .
