Agar vs. Liquid Culture 1

I have read up on the use of agar and liquid cultures. I was wondering if one of you experienced cultivators could explain the purpose of using agar and maybe give a compare/contrast of agar vs. liquid culture. Any reply is appreciated. Thanks.

I only use agar for isolating strains, liquid culture is great because you can make 1-2cc's of spore solution into 3-4 cups of mycelium soulution, you also dont have to wait for mycelium to germinate. When I do agar I use the wedges to make LC's

LC is practical in that you can turn a few cc's into hundreds with a little bit of research and practice.
Agar is a not as user-friendly as liquid culture, and is used more for isolating and cleaning up prints. Not really necessary to use if you get a vendor syringe or print.
You can get away with never learning agar work in this hobby, but LC is a must IMO.

contamination is much easier to see on the surface of an agar plate than in Liquid culture. tissue can be cloned directly to agar, whereas using a chunk of a fresh fruitbody to inoculate a liquid culture has never been successful for me. you can muddle up the mycelium on an agar plate with the tip of a syringe filled with sterilized water and suck up the mycelium... this works better than innoculating with wedges for me as it gives more inoculation points and you dont have to open up the jars to get the innoculant in there (less chance of contams)... you'll need a decent glovebox to do agar work successfully, but i feel that working with agar is both fun and rewarding.

is agar work worth learning as a novice with only 3 grows under the belt or should i wait until i am more familiar with the growth and habits of the mushroom? Agar work has always confused me...

It still confuses me, and I'm somehwat of a n00b like yourself. I'd recommend Agar's baby bottle tek, seems very very simple.

-Most agar work will need a glove box. LC's do not. -Agar needs... agar! which some find hard to get. -Agar needs petri dishes or low jars, also hard to find. -Agar lets you see growth in "2D" so it is easy to see if it is rhizo, and if it is contams. -LCs are dirt cheap and simple to make, you can suck up using a big 60ml syringe and inoculate lots of jars with no need for a glove box. Agar dishes will need sterile transfer to all the jars. Only so many wedges per petri.

Agar allows you to isolate individuals from populations in a controlled way. Liquid culture allows you to rapidly expand that individual, or populations of individuals.

My only attempt at agar failed due to contamination but that was years before I found the shroomery,I look forward to giving it a try again in the future now that I know so much more than I did than

Agar will prove very useful no matter where you get your prints/syringes. Every time you do a multispore inoculation numerous substrains are created. Doing agar work allows you to isolate a substrain with positive growth parameters like rhizomorphics growth.
