My first time doing agar and i do not think i am succeeding. I streaked a couple plates and did a transfer from what i thought was something healthy growing, but i am not sure i did well.

Here is a B+ i did but it looks like fluffy mold:





Here is a PE one that i think is doing better, but it could just be all bacteria. I dont really see any good mycelium so im not sure about it:





Heres another PE sample that looks pretty bad. There is one area circled in red that might be ok to transfer? or maybe that is bacteria too.:



It looks like there may be something i can salvage from one of the PE ones, but i am not sure.

As long as there aren't any visible contams, it's fine! Perfect world scenario, transfer before it reaches the edges of the plate, solely because the edges could potentially harbor a contam that isn't apparent. About dense growth, one of two ways. Either get lucky from the print and get thick growth off the bat, or transfer a few times until it thickens up. I had a culture that after 7 wispy-ass, thin-ass, weak lookin plates, bam 8th one was rhizo as hell!

Yeah the potato pics make it difficult. The first two PE look pretty good, but clearer pics are needed.

Using a print is MS

MS= multispore (lots of strains)
Vs
Isolate= single strain

Did you mean syringe?

Prints have more spores condensed and easier to see on the plate when doing a swab. Syringe is significantly less spores suspended in who knows what if you didn't make it yourself. Plus as a noob you don't run into the issue of putting too much solution on the plate. There are other benefits to prints but I am biased towards it. Not many seasoned mycologists on this forum use syringes unless selling them or they are filled with liquid inoculate.

I tried filtering the potato bits:

  0730 10g potato, boiled for 10 minutes, then 8g agar, 6 drops red.
  0750 coffee filter failed, it collapsed over the edge
  O755 again
  0805 failed, just potato, but clogged filter,even using a pre filter
  O806 again
  0820 50% potato, just going with whatever!  :shrug:  8g agar, 6 drops red, to just boil.
  0830 into Presto 23 Qt, 10 min vent
  0848 25 min PC
  0915 PC timer up, turned heat off
  0945 30 minutes till valve dropped, out with media bottle and tightened cap.
  1145 Agar @115 F

I can't wait to get some Lite Malt Extract DME.
Mebbe Briess, anyways everything is hard to get here.
But I may have found a brewing supplier here.

So you tried just filtering the potato juice (before agar)? That works for me when I want to be anal, but I use potato flakes (instant potatoes) when I make PDA.

That's the right stuff, the only thing I do differently is I run my flakes through a food processor first to chop them into dust.

I'm a newb too and i find that using syringe is messy for me also. Use sterile swab to nocc up with syringe. With that said my germination rate with prints is at about 70-80% right now, much less bacteria than the syringes. Only thing you may need to worry about is mold. Transfer early and youll be fine. My advice if you make messy plates with the syringe, use a print. Worked wonders for me.

Good way to utilise a syringe onto agar is put a drop in a sterile shot glass then use a loop to streak the plate. Works well for me and avoids all that liquid on your plate :thumbup:

Can someone explain to me the connection with crazy looking plates from spore prints... and how using too many spores makes the mycelia super fluffy... and takes numerous transfers to even it out?  Whats going on with that?

So, here we have a bunch of different strains fighting over the agar.
It's chaotic and there's no fluid movement.



And, here, when we've isolated a strain, we see that it can take a pretty even course forward with the agar.
