Re: AGAR ENVY! (Anything and All Things Agar!) [Re: Edmunter]

Hey guys... long time grower but started going a bit deeper and doing agar work the past few months and I've kindof been coming up at a loss. So wondering if anyone has seem something similar or has advice.

Here's a PDA plate of a Cube GT after 1 transfer:

I've seen these droplets in multiple plates I've done with various syringes (both cubes and pans). Everything was done in a lab setting for max sterile technique. I've searched posts and see one or two about metabolites and then some about mycogone/vert contam but I didn't think that was possible starting on agar.

Any thoughts? Please direct me to the correct forum (thought maybe contam)? But since this was agar...

Okay that makes me feel a lot better because I couldn't think of a way I had that many contam'd syringes or I screwed up THAT bad but was still scared. lol

I know it usually gives off heat as it grows but does that potentially mean I'm keeping my plates "too" warm as well? Or it's just doing it's thing?

PESA MS (germination plates)
Malabar clone
2 Costa Rican clones (Tall/Meaty & Short/Fat)

Anyone here ever use black food coloring?

How did you like it?

Did it make anything harder to see?

How dark?

I used black tea, made the agar black and apparently has anti bacterial properties which I seem to agree with so far thereโ€™s a thread somewhere for t-gel agar

What are you guys using to get nice round agar transfer? I was looking around on amazon last night and two options I came up with were hollow steel leather punches and dermal punches with plungers.

If using dermal how are you sterilizing them? All the ones I seen were plastic disposable with SS tips

I know some people take large gauge needles and poke them unit the agar so a piece gets inside then push it out into the new plate.

Why do you want round transfers?

Those look good to me.

Iโ€™ve never used the multi compartment dishes but I would either do like you said and use half for say T1 then the other half to move a piece over for t2.

Or maybe take a T1 on each side then pick whatever looks best to expand from there.

The divide goes to the lid right? So if you used one side and got bacteria or something you could take a quick transfer over the wall and not have to use a new dish.

It looks like mycelium can get over the divide fine, but I am not sure if bacteria will be able to.  I am thinking about leaving this pate be and taking a transfer once it hits the media on the other side (it hasn't quite yet).  My thoughts are that this mycelium on the new side of the agar must be pretty clean.

Anyone have thoughts on this?

Oh thatโ€™s crazy. I wonder how likely it is for bacteria to hitch a ride over the wall on the myc.

I think this is my new favorite color.

I just remembered reading about a technique to get clean myc off dirty plates where you put another chunk of agar on top of the myc because it will climb up to the new piece and leave the contam behind. Seems like that could be one of the functions of the divided plate.

It still seems like spores or bacteria would be stuck to the mycelium and travel with it but I guess not.

Bacterica on Agar due to poor sterile techniques.
The Bact is keeping the far right myc from growing at all.
The Center myc has setup an intransitive barrier.
The funky bald spots is from dripping drops of ISO after the transfer. Wanted to check bluing for kicks.

Aesthetics, Iโ€™ve noticed the plates that have really caught my eye all had perfectly round centers and the myc spread out uniformly from the center.

I'm sure you probably know this already, but you don't necessarily need circular transfers for circular growth:
