Anyone know for sure what can cause agar to not set? I had a couple batches that didn’t solidify. I’ve heard ph can play a part in that but it seemed so nuts that 2 in the last 50 batches didn’t set up. After last night’s fail I did another batch that I just cooked hotter and longer while mixing and it was fine.

I’ve been trying to wait these out but it’s bugging me.  There is a bit of condensation but think the pic is clear enough.  This is APE.  My first time with it.  Does it usually look so grainy?

Do you have too much agar powder? Less makes it harder and more makes it softer I believe

I use 7g agar, 7g MEA, 500ml water. It went runny twice prepped with red food dye. It worked like it usually does after I didn’t rush the process and used blue but I don’t think the different colors have seriously varying ph values or anything. Only happened once before but I may have been rushing each time. Seemed weird bc it always sets up but last night I was cooking agar while setting the heat for pc grains and also spawning tubs lol.

Your ape plate doesn’t look bad. Albino varieties in particular can look a little funny.

2 tissue samples taken from the same fruit from RW clone #1 (hence 1.1&1.2):



Will be taking transfers from these guys after dinner... Grabbing the 5 biggest rhizomorphic ropes from 11-1  in the PE6 clone culture I pulled a transfer from out of cold storage since early june... taking 5 transfers from RW between 5-9, and ideally 5 from red boy from 7,10,11,2, and 3... second picture is APE T1 going rhizo I'll be getting 5 from between 1-6, and the last showing a close-up of the red boy with some crazy sectoring:



3 T1 transfers from a clone, from RW clone #2, looking clean, may go to grain:



Cultures that will be split to 2 jars a peice in 3 days  (boil oats tmrw, let dry overnight, pc the following night, and A2G drops Saturday morning) will be comparing tomentose and rhizomorphic PE6 cultures:



2 AA+ plates (T3 and T4) knotting up:



PE6 MS tub >clone>agar pin clone >agar pin clone, and the first 2 pins from it... so 2 3rd gen agar pin clones:

Exactly the opposite, more makes it harder and less softer. 9g agar to 500ml is my go to, with spores its 7g to 500ml, they like a little more water.

Thats funny because i was going to also make a comment about the red food coloring completely gone from some of my agar plates.

I only colored a few times... found it to actually make it more difficult for me to spot any contaminants

Did you have any hard sediment? Perhaps it was not mixed well?
10g/500ml is the usual recipe, at 7 you would be on the soft side. Which recipe are you following?

Iv heard of people putting the agar/nutrients in the flask and then dumping h2o on it, not mixing it at all and PCing it with success. For real the only reason you get soft agar is you don't have enough agar in your mix, maybe they were a little lite. If you mix 9g agar to 500ml h2o guaranteed you get firm agar.

Did you have remnants in the vessel? Like left overs that set before pouring?... I put my dry ingredients(3g agar and 6.5g LME) into the vessel, put roughly 120ml of hot water from the tap and swirl it quickly before the malt clumps, then fill it the rest to 500ml with hot tap, pc directly... I still get sediment, but not bad

I always cook mine for 5min or so, it would be alot easier to mix it in the vessel, I always have sediment in the agar, never worried about it.

Caps, loving your info man!  You've been a big help to all.  Quick question, are you working in a SAB?  I have an opportunity to head to a lab with plates and agar in a flow/fume hood workspace.  Do you think pouring and transporting to my home mini lab presents an issue?  I'd like to take the opportunity to do some transfers and pours in front of the hood but can't leave em sitting there once done and set up.  My SAB is working but I am having contamination issues in places.

The reason I'm doing it (I didn't start doing it, I started dye free) was because I was having a hell of a time finding the vanguard of good myc on the surfaces I work on.

My tables are all woodgrain, which makes finding the edge of growth pretty challenging. So I started to add color to differentiate from the texture of grain-y-ness better.

If I had a proper lab and shit I'd probably not be using food color.

I have a roll of paper towel handy at all times and set plates on that...I too have woodgrain workbench made from a solid core commercial door.  The paper towel is white as pure snow and helps with lighting as well.
