LOL well I just didn't want to get into the details. And shit, my job don't need an explanation for what I'm doing, I almost never give them deets :P
And I will do. Honestly, I'm just messing around with recipies. I've had success with everything I've tried to varying degrees. I'm guessing that, while it would perform like crap, you could probably just do straight water+agar and you'd get colonizing, just really light.
Mostly with this attempt I want to give the myc a better surface to latch onto. I found with the last plate set that if I cut v. small segments that occasionally the myc would just like... slide off the agar (too firm).
I think I was overall most afraid of agar out of all the things I've learned about mycology, and then found that it is the most forgiving part of the whole process.

I couldn't agree more about the agar thang! My last plates were poured hot and three of them not allowed to set so I have three plates of cell soup. I did a 12-14% PDA concentration like I was setting up agar for a solid gel electrophoresis...turns out it's a bit rich and my samples went translucent, but they are starting to 'fuzz' out nicely. Love the agar, can't wait to start coring and transferring to new plates and/or grains...takes the MS syringe gamble out which I like the most!

Yeah, this exactly! I spent 3-4 months trying to get things to work, having everything as clean as possible, then BAM 2nd flush green in the tub. Or pink. Or black in the grain. Or orange... SIGH.
Now, I got my contam down to just happening in agar if I do enough transfers!

First agar plate here but I've had previous agar plate experience just not with mushrooms. It wasn't great but I think it worked out, from what I've researched at this point I could start grabbing sections and transfer these onto a new plate correct?
Also on the third plate there are yellow growth below the white, contaminated I'm assuming? Pictures could be better but that was the best I could do atm.
First two pictures are the same plates. Third picture is a different plate. 2 plates total. Ignore the bubbles, I rushed it and the agar wasn't set all the way before I dropped some spores in it. Also too much condensation on top otherwise I would've gotten pictures from the top.

Transfer once the culture is dime sized, take small (rice grain) transfers from the most uniform leading edge
The yellow is bacteria, bacilli I believe (not that it matters much) transfer away from it... anything that's not white mycelium is contamination, and some contaminants have white mycelium as well...

I'm really terrible at this because it's soooo hard to see into my SAB. Any tips on getting smaller transfers? I end up around 4-grains-of-rice-ish when I try really hard to "go small."

That looks really awesome... but a glass plate... near my wrists? How do we get away from slicing off limbs?
But seriously... that looks like a great idea, totally nabbing this. I was peaking through my damn arm holes last time and the angle I had to hold myself at was making me sweat after a couple plates.

I look through the armholes... you can use an inoculation loop for small transfers as well... I've not been using my scalpel unless doing A2G transfers

I will be attempting agar in few days and wanted to ask before I get started. I will be using a MS Spore Syringe. I'll be working out of my SAB. It should be ok to just drop 1 cc of spore solution unto center of agar plate? The sides?? Also, how long does it take spores to germinate on agar?? Was gunna use colonized agar to do G2G Transfers, if they colonize that is

One drop only... shake, flame(both outside of SAB). Once in the SAB (immediately after flaming) run one drop to cool, open your recieving dish,and while holding the needle nearly horizontal (to avoid hovering over the dish) drop a single drop of solution near center... you want the least amount of fluid on the surface of the agar as possible, as it can run and spread contamination in an otherwise manageable situation

Perfect, thanks for that. 1 single drop. Man, is it safe to use agar with spore syringe??
What is best way to germinate spores? I hear alot of bad things about syringes. Like, you shouldn't use with grainbags, it seems like BRF is only good to spore syringe. Is this true with agar?

It's the only safe way IMO... use agar to make sure your culture is clean: it's a good idea to wait until it's dime sized to confirm it's mycelium, and to make at least 2 transfers out, whether it looks clean or not... best to burn up agar supplies in a matter of days/ a week, then to be a month+ into a grow and hidden contaminants rear their heads...

Yes I agree!! I was just hoping spore syringes were OK for agar, especially since germination can take time- also since it seems like they are not recommended for anything except BRF.
