Take your largest,fastest finishing fruit from clusters for clones... use the weight of the smallest of those as your test dose, as you won't be able to bioassay anymore of that particular fruit than you have... i.e., if you have 2- 2g fruit, and one that's only 1.5g, dose at 1.5g to get a more accurate comparison... you can't take 2g of a fruit that only weighs 1.5g

I pour my agar soft as hell so hydration isn't necessary... the print you'll receive in a few days was printed last Tuesday, so shouldn't be any issue anyway... only use a small amount and you should be able to stretch the print to printing your own in masses

Ya, worded that wrong.  MS solution=hydrated spores.  Haven't gone straight from spore print, so wondering if hydration is necessary.  From what I've read up on here, no.  But, still wanted to double check. hate to waste a dope print:)

You should really always have spores for RW now... it's not a single use item... there are millions of spores there, only takes 2 in fact, I've given away 30 swabs from the F9 print I recieved, used it 3 times,  and still have well over half of the print

What is difference between strain isolation and cloning?? Will strain isolation provide equal genetics like cloning does??

Strain isolating is v difficult from what I understand. What people tend to throw around, around here as "strain isolation" is still pretty unisolated. It's effectively trying to narrow the different strains within a culture. So, if you put something on agar and you grab a tiny tendril of mycelium and put it on to new agar, you're "isolating" further. It seems likely impossible for the amateur mycologist to actually isolate a single strain. This isolation can make it so that your tubs of bulk fruit at approx the same time, look similar in shape and form, and have similar potency.

Cloning is putting fruit to a substrate where it breaks back down into mycelium that you can then do strain isolation from.

Mhh, I just did my first two inocs on agar.. because I have so many plates and I don't feel like waiting for a fruit to clone.

My hardest thing is just getting a single fucking drip out of the syringe. I had a little "shot" come out on both the plates I did. I hope that doesn't fuck anything up :/

I want uniform flushes and fruit size, etc. This can be done with cloning tissue or will I still need to isolate further??

You could isolate a shitty strain... starting with a clone guarantees at mimimum, a culture that will fruit... you can do further isolation, but I've found original clone cultures outperform 11 of 12 isolates derived from that culture... a clone culture, though still containing multiple strains, is a monoculture, as it works together to perform as a single culture... but without

Thanks again caps. I will not worry too much about isolating as I have done 3 transfers to agar and myc appears clean. I will put those to grains and then monotub it. When fruits have been produced, I will clone the tissue and further the agar from there. I just didnt know if I should be wasting my time isolating from MS, if it is cloning I should be using for uniformity.

while a noob to this forum, and to the hobby, I have 18+ years experience with syringes and exact med pushes.  Here's a trick, apply counter pressure, or "traction" on the plunger.  Hold the syringe in your fist, needle down/plunger up, with thumb up and pinkie, ring, and middle finger around the barrel of the syringe.  Take your index finger at the base of the plunger and squeeze while you apply down pressure with your thumb.  Play with a blank water filled syringe for a bit to get the pressure of index finger vs thumb correct. Kind of like finding the friction point on a clutch...once you've got it, you've got it.  This will help when you get a clog from spores so once cleared it doesn't 'shoot' out. Conversely, you can do the above but rather thanking thumb on the end of the plunger, pinch the plunger at the base with forefinger and thumb and you have more control over how far you are pushing the plunger...if you need I can upload a couple pics to clarify...happy to in fact.

You'll get uniformity with isolation,  whether from MS on agar, or cloning ... just may end up with a culture that uniformly sucks, lol... at least with clones, you know it'll fruit

LOL yea! Thats whats worrying me, is getting a strain that sucks ass, or low potency. I suppose cloning several pins or tissues instead of one then fruit em out and see the best performers

I grow MS as genetically variant as possible  (basically T2 if it's clean)  and pick the 5 fastest finishing from the largest clusters ... clone mature fruit so you know they finish well... after bioassays,  I pick 5 fruit to clone from the winning culture's tub, and keep the parent dishes until these have grown out and been tested... if none perform well, go back a generation  (grandparent plate I suppose lol) and try again from the culture you know had the best genetics of the original 5 cultures

Hey thanks for that man! That'll actually help me a lot! I'm a noob to it all still. Practicing. I have a blank syringe I will start practicing with. If you could upload those pics I would be highly appreciative. The more I know how to do things more efficiently, the easier this hobby will get.

Here you go diggity dog!  A couple different hand positions for you to try out.  In my SAB I usually do the pinch method due to height restrictions, but you do what works for you.  Better to practice with blank than toss spores, LC, or anything else tasty away.
