70% Iso. Alcohol - Sterilized too well?

I figured I'd start practicing some sterility after an average of 1/4 of my jars were getting contams... finally got some tyvek and alcohol HOPING I could put those days behind me.  :tongue:

Well, after inoculating 10 1/2 pint jars of BRF with Keeper's Creepers and and 6 quart jars of WBS with Gulf Coast, I think the alcohol may have killed every damn spore coming out of the syringe. I simply soaked a paper towel in the 70%, and laid the needle in it whenever I was taping holes or doing something else etc. Now, 6 days later, there is NO growth and ANY jars. I've never had that happen before. They are at perfect incubation temps, and both the BRF and the WBS was PC'd, 30 mins and 60 mins respectively. 

How exactly are you supposed to use this stuff without killing your spores??  :evil: Is it ok to re-inoculate jars that didn't receive any viable spores to germinate? As far as I know the jars are still sterile.  :rolleyes:

wipe everything down with it and then give it a sec to evap...or blow on it....

Hmmm I lay my needle in a cotton ball soaked in 90% ISO with no problem.

blow on it?  Tons of bacteria in your mouth...  don't know if that's such a good idea. :frown:  maybe use an asperator instead?  Like for cleaning camera lens.

blow on it??? Wouldn't that defeat the purpose of sterilizing, seeing as our mouths are filthy??

Yes blowing on it would probably recontaminate it.

You should wait atleast another 2 days before you reinnoculate IMO just incase you are having bad luck and they are about to start growing.

blowing on it....fanning it in the air....my friend never had a prob and never has contams....find what works for you


oh he said he likes to gargle the iso alk for a minute then he blows on it... :doggystyle:

If you just wiped the needle down with an alcohol wipe like everybody else does, nothing on the inside of the syringe would be comming in contact with alcohol.

If you havn't worked with this strain before give it time. It can take longer sometimes. I have a strain going now that took over a week going to show any signs and one still hasn't (as of 3 days ago on day 10, havn't looked yet, will this afternoon) but I'm confident it will.

It's highly unlikly that the alcohol killed it..it evaps very quickly and if it hadn't by the first hole, it would by the rest of the holes. Not to mention like discman said..very little of the inside of the needle is sterilized with the alcohol (a little will get in the tip sometimes). Thats why we flame sterilize as well..

You also shouldn't need to force any air over it. Just wait a second..it only takes a few for 70% iso to evaporate.

You should have a paint mask on anyway when working with your shit so you can't blow on it :wink:

I do the same as you discman when I don't have my medical mask on.

Sometimes I find myself holding it until I feel like I'm going to pass out.  :lol:
