best way to sterilize needle?

all i did was this on my first set of jars. i turned oven on to warm pulled rack out sprayed lysol(and of corse u have santized your area with lysol and alcahol) i flamed my needle with a lighter till red hot then i wiped it with alcahol to cool it i injected my jars 4 holes. i put syringe strait through tape on holes i resterlized with lighter and alcahol every jar to avoid cross contamination i did not retape the holes i covered back with foil then i put in incobation chamber(aquar. heater set up) and that was like 8 or 9 days ago. i got my first pins today on one jar of lipas and the pesas are not far behind Peace and may the force be with you -Lee

hola matt, what i usually do is i sterilize the needle by makin it red hot with an alcohol lamp or a butane lighter. i find the easiest way to inoculate is by turning the stove on and pulling out the bottom rack as far as it will come out without falling out. use this as your platform to set the jars. vthe heat from the over should keep all the bad contams away. hope i helped. adios. peace.

Your needle should already be sterile if it's coming out of an unopened pacakge. And after innoculating the first jar, it should still be sterile. After all, your jar shouldn't have been contaminated so neither should your needle. All this flaming and rubbing with chemicals, its a bunch of overkill and shouldn't be needed. It only takes a bit of common sense to be working in a clean area.

same here you can get a pack of 100 alcohol wipes for a couple few bucks

Agreed. I recommend inoculating over a warm oven door, and make sure the isopropyl alcohol you use to wipe the needle is at least 91%. Peace.

Actually 70% - 80% alcohol is more effective at killing germs. The higher percentages make for better flames...

When using the oven tek, and if you have a gas oven, just turn on the front left or right burner and youll get a nice hot flame to sterilise the needle with quick. Wipe with alcohol to cool. Im a clean freak and heat the needle after every jar.

those suggestions helped greatly, i wiped with alcohol and heated before every jar over the stove; i innoc yesterday, but ive been wondering how much moisture should be in the jars? mine dont have any! peace Even castles made of sand, fall into the sea, eventually

might want to be careful spraying lysol near an open flame (i.e. the oven)!!

I had B+, thai, australian, and equador eq got contam, thai got contam, aust didnt do anything, B+ took off like a son of a who re all on PDA agar inoculated some more pitres with the 3 that didnt do so well the first time. Funny thing iwht them though, they worked the very besdt in the brf jars oh well. B+ is a fast mother fucker when it gets going on agar like 2 days, and a .5x.5cm slab of agar will colonize a new pitre. if placed in the middle.
