Update on this weirdo- I'm really curious to see if this fucker pins! Also a pic of a shoebox that's going nicely. The shoebox was from one transfer later (I don't know if I label correctly- from spore that plate is "spore", and the first transfer for me is T0).



This cluster has a similar alien look at the base, it was closer to the original plate pic before the cluster formed.

Why, if I may ask? By making more transfers, you just decrease the genetic diversity, so personally I take the least amount of transfers possible. That way I wonโ€™t discard the best genetics from the first clean plate, and I can clone the fastest or biggest fruit, or the nicest cluster. At least thatโ€™s my logic, but Iโ€™m not an expert.

Really only kuz spores are dirty and to clean it up. So I usually take a few different transfers from different areas to get some genetic diversity because I usually dont like keeping the original spore plate unless it's super clean looking and even then I end up cloning or printing from a fruit I like. And if one makes plins then even better.

Sorry for the second photo being so shitty. BBBBUUUUTTTT so excited had to share.

Iโ€™ve had such shitty luck with preserving TWAPE. All my swabs came out contaminated af. So resorted to BRF to get something to clone. This clone transfer was so shitty. I was so frustrated I left it and forgot about it and saw this tony spot of mycelium threading at 7 oโ€™clock I wanted to see if I could possibly snag it and keep going.



And hereโ€™s the transfer 3 days later!!

Hi!

I just started dabbling with agar. Created a few from the LC I bought and also transferred a couple that looked contaminated.

I want to ask you guys if these look healthy to you:

This was transferred from another one as it got contam. Is the lower part fine, or should I be concerned?


Lion's mane



Made from an LC by just squirting into it. A bit all over the place. Should I move this into a clean plate, or will it continue to colonize? It's stuck like this for a while now.


Shiitake



Very slow growing. I think it hasn't changed a bit for a while now. Should I do anything with it?


MDK



Some of the spots are cotton-y looking. Should I do anything with it?

Some gnarly looking plates lol. Are these all freaking out over some nasty contam? These were started with pieces of a swab stuck into them, kind of a fafo thing I did, so nothings riding on these. One of the plates I streaked with this swab finally germinated the other day and I got a nice transfer from it ๐Ÿค˜

Recipe seems off based on growth.
I made some PDA plates using 500ml water, 100g of boiled potato water and 4g of LME. After putting some LC to the agar the growth seems stunted and only growing upwards for the most part. Mode like cotton balls growing up and not spreading out looking nutrients. Too many nutrients in the recipe? I did purchase some LME, peptone and dextrose premade ones that are looking like expected growth.

The first 2 are the PDA plates I made



The last 3 were the purchased plates

There's probably a bacterial contaminant in there if that's all the growth from a transfer after 25 days. Cabin sequestration or trenching might help clean it up

So I have a question. This isn't the first time that this has happened, where putting colonized grains on agar results in VERY rhizomorphic growth.



Now, this makes sense. The mycelium on grains was adapting to colonizing grains, millet in this case, and are in rhizo mode, spending energy to travel up the jar and break the hull, etc.

Compared to the grain, the agar solution is low nutrient, so it will continue being rhizo as it expands outwards, right?

Does this also mean that the mycelium that went from grains to agar already has the enzimes in its arsenal to digest the grain at hand, therefore improving colonization?

Let's say I swabbed something on agar, made a few transfers, but this mycelium ONLY knows whatever Agar recipe I had.

When put to millet, it will have to learn to produce the enzymes required to digest it, right?

Well, if we take that colonized millet, put it back on agar, then would that mycelium perhaps colonize faster?

Has anyone ever measured the jump time from agar to grains before?

whats the best tech when we do transfers in agars!? the litle part that we take from old agar, when we put in new, its better: - put with part mycelium down..touch base of new agar; - or put the part of mycelium up..??

I've always done mycelium up though I think it's entirely preference of the person doing it.
