well, i've seen other threads about LC and some you had commented that you use LC without a syringe, just pouring the LC in grains like G2G transfer. for my surprise, this seems like common practice with LC for some reason, i didn't know that.

my idea would be using syringes instead of doing it that way.

i've seen that doing the "no gas exchange LC" with syringes could potentially create a negative pressure inside the LC bottle, that could suck air inside the LC and potentially contaminate everything.

what are your thoughts about this? i didn't want to abandon syringes because i find them very useful and less contam prone than just pouring LC, even if i've seen some TC saying otherwise. i use a SAB instead of a flow hood.

the negative pressure inside the jar is also a genuine concern or definitely exaggerated?

Firstly... Why do you want to run a LC without a gas exchange?

I believe pouring is better because there are less vectors available to spoil everything.

How much experience do you have and how are your agar skills?

guides like the mushboy guide to liquid culture also say that you don't need gas exchange using a broth. using micropore tape for gas exchange isn't foolproof, because some bacteria can still get through, and the tape can get wet and become a vector for contamination, and i didn't want to buy a 0,22 micron filter or something. hard to find where i live.

theoretically, using a syringe would be better, at least that's how i see it. a SAB isn't sterile, but a syringe is. using a syringe means taking material from a sterile jar (the liquid culture bottle), transferring it into a sterile syringe, and then transferring it to sterile grain. that seems like the safest way, with zero contact with potentially contaminated external air.

zero experience, first time doing it. feel free to correct me, i'm here to learn.

When using a broth with 0,1-0,2% DME(dry malt extract) the food will be gone before the available oxygen, even with a closed off unmodified lid.

Free pour is safer as long as you don't reuse the LC. One bottle for one session and why I suggested the 100ml bottles.

If you plan to make a lot of LC and keep coming back to it over and over again I think a modified lid is better. Have a look at the Dreamer bottle as well.

If you include why you want to use LC in the first place that would help narrow down what solution might be best.

Honestly as someone who’s made and used gallons of LC I can’t recommend it for casual use. Unless you’re growing many pounds a month or working with a specific species that benefit from LC expansion, I can’t recommend it. Is definitely a higher skill set and if you’re looking for shortcuts in the shortcuts, I can’t see it working well long term. To repeat DV8DUG how are your agar skills? I would never inoculate any LC with anything other than clean agar wedges.

I totally agree, I only use LC for slow moving species that don't really like grains all that much.

I haven't messed with LC but do do Rev's LI tek, mostly for maintaining cultures but also to inoculate. I just got a bunch of 100ml media bottles to preserve a new batch of cultures and won't be making LI, just preserving. I get full colonization from an agar wedge within a couples days of what I get from LI. In these warm months it's not uncommon to be all white in 9 days from a wedge after a couple of shakes.

So yeah, all the potential downfalls of LC just aren't worth it to me, doing ~15 shoeboxes a month.

I can testify to this.  I decided to try it once out of curiosity, because why not?  I was successful, but I just didn't need that much.  The most I've had going at once is 2 shoeboxes and 4 monotubs, and even that was easily achieved with a few clean agar plates.  When I made LC, I had so much excess LC that it just felt like a waste for what I was doing.

For me, the single biggest factor that made LC an "advanced skill" was being able to identify different bacteria and contams and stacked scenarios/vectors in a sometimes cloudy 3d media, (SSLC) rather than a simple, clear, 2d plane. Regulaar LC should be pretty clear if you make it properly.
There's just exponentially more that could go wrong. Coupled with the amount of time and energy you put into expansion, and the risk bottleneck you take with something as delicate as LC that hinges months maybe years of work and your entire workflow/current grows..
cost of diminishing returns, cost of hubris, icarus etc.

I have a question? When I’m taking prints in front of my flow hood, do I need to leave it on for the 24 hours while they drop? Or can I cover them with a shoebox/tote and turn the hood off until I want to lift the shoebox/tote again? I can leave it on, but it’s a magnet for my cat and I’m afraid if she will contaminate the shit out of my prints, and rub up against my filter until every plate I open gets filled with cat hair and bacteria. She loves me like that.

Proof of an 18 year old cat contemplating the biggest jump of the last 10 years with no place to land, just to contaminate my shit.

I'm not sure leaving caps in front of the hood would be a good idea even without a lovely cat like the one you have. Caps would dry out quick and unless you make a single line with the caps whatever is on the caps would just blow around on your foil.

I like using some sort of tote for printing. If I'm printing something that takes time I'll put plastic shot glasses over each cap.

well, i want it to inoculate grains easily and safe. i already have 100ml of liquid culture, i want to multiply it, to have more and be sure i have enough, for worst case scenarios or something.

i will do a modified lid with a SHIP to use syringes. just wanted to know if it was better to open another hole and use micropore or just the headspace was enough.

i also can't understand why pouring would be better, in any scenario.

for example, when using syringes the workflow is like this:

"liquid culture inside a sterile bottle > pull inside a sterile syringe > push it inside a sterile grain jar"

everything is sterile from start to finish reliably.

if i'm not mistaken, a SAB is not sterile, just reduces contamination.

so, pouring would be:

"liquid culture inside a sterile bottle > open it to non sterile SAB air > put it inside a sterile grain jar"

which introduces a vector of contamination. with agar, this vector also exists, as you need to open the agar plate to the air inside the SAB. that's why contrary to what pastywhyte thinks, i would say a LC would be a better inoculant if it is verified with agar before inoculation, to see if it is clean. then you can simply use it reliably using sterile syringes in several grain jars with very low chance of contam.
