You're already on aliexpress. Buy the $15 impulse sealer.

I heard that twistties or cable ties are an option, but they seem kinda risky imo

Is there a way to get spore prints or swabs from yeti?

Absolutely, swabs are gonna be the tool of choice for this one’
Let em go for an extra day when you think it’s time, they will stretch and pack on some weight. (After 1st flush)

Thanks, that’s what I figured. I’ve cultivated them a few times but want to stop transferring and making new plates. I just want to preserve the culture for future use.this was a tub from last November. I’m guessing this was about the time you are referring too. I harvested these the day after I took this pic.

I'm sure this has been asked before though if already having sterile receiving container with lid In a still air box would it be possible to make a lc broth and sterile-filter all contams out with a very small fine micron screen n all clean screen-sterile lc into receiving container to inoculate it later with mycelium

If you use a filter capable of filtering bacteria it won't allow passage of the mycelium, it will clog the filter. You could probably pump uncolonized broth through a filter but it would take a while. Steam sterilization is the easiest route for broth. Infected/colonized broth can't be saved by filtration as the mycelium is larger than the infecting organism.

If it's just bacteria in the LC, I've had success with a BRF puck. You can fruit it for a clone or spores, or just let mycelium grow a bit and transfer a small piece to a water agar cabin sequester. That will get you a clean sample of the genetics so you can make another LC or just use the agar.

Re: Ask Quick Questions, Get Quick Answers [Re: tripdawg420]

Forgive me for asking on this thread I tried the agar thread and wanted some more answers. For context: I believe it was may 7th when I made my first no pour brown rice water with corn syrup agar in some 6oz mason jars. I put some Jack frost spores some PE 6 and a tissue clone from blue magnolia. As of today the blue magnolia is doing great even after multiple transfers were taken from it. The Jack frost hasn’t been opened yet but is doing good as well, clean circular growth and the PE looks probably contaminated which makes sense because when I did that I didn’t sterilize my swab I used because I didn’t really know better yet or think to, my mistake. My next two batches of agar were with just corn syrup first batch with dye second without. Did some transfers and dropped some liquid from vials and even put the agar to mine as a transfer. I’ve been very discouraged and frustrated not knowing where I’m going wrong and how to go about fixing it. I received a nice amount of cultures which I do not want to lose. On another note tho On the 28th of may I used swabs i sterilized to swab some ochra on two plates. (Thank you archespore) as of today I see growth from those streaks. Here’s a very shitty picture which happened to be the best one I could take I apologize
I also used some swabs that contained spores already on the say day (5-28) of some Australian landrace cubensis and I don’t see any but that’s not a big deal except the ones I put to agar on the 19th aren’t showing either. Not a huge concern but I have a bigger concern about my other cultures. A thought I had was that the first batch I used was Much much harder than any of these batches I made since. I’m wondering if the softer agar has reason to do with the mycelium seeming like it’s growing inside and under the surface more than it is on the top. I see very whispy clear almost growth and closer to the actual transfer are circles most agar plates tend to have when their growing out so I’m confused. Anyways. If these cultures ARE contaminated and it’s not my agar recipe, what can I do to save these cultures if anything at all. Here’s some pictures the best I could take.

I put my biggest tub out to pasture today, so I only have a few small tubs left so I really want to get some clean agar to throw in some grains asap as you all know this hobby can keep us sane and I’d be lying if I didn’t say that this is causing me a lot of worry and frustration so any suggestions and solutions are greatly appreciated. Sorry for such a long post

Ok. Sorry. I’m trying to follow along with your post. It’s kinda confusing.

But first

1) you can use a sterile swab. But no matter what, what you swab is not sterile. So every culture is “contaminated” and needs to be cleaned up

2) Did you say you’re streaking the plates?

In all honesty I find streaking to be a waste of resources. One of the best advices I got on this site is to take sterile tweezers and rip the swab off and bury the swab in the agar. I get growth and if the recipe is good you’ll get multiple transfer points to clean up from

As far as the plates go, the sixth seventh and eight pictures have good spots to try and work with

3 Months ago I found a 7 year old envelope when I was doing inventory around my place looking to setup a myco work area again, an unopened envelope with 6 different strains each strain had 2 swabs very neatly and profesionally packaged. Streaked all those onto 30 some plates and a month later only two plates out of 30+ germinated and it was the same strain PE I was dissapointed to say the least.

A month later I used the darkest looking swabs and really dug into another 20 plates or so, and a month after that NOT A SINGLE germination, no bacteria, no NOTHING

Can someone plz comfirm for me if 7 years is too old for spores to germinate "normally" like fresh spores? I've never tried germinating anything that was older than a year.

Also these plates of mine were made from several different batches of 2% strength Grain Water, BRF, Honey, Cat Food and MAE, and a batch of 4% MAE, so I gave these spores more than enough and 3 months later only two plates germinated :begger:

@awfullotofshrooms I think pretty much all of those plates are good just let em grow out and note color and growth, but I see a lot of mycelium. I'm dealing with the same thing right now, I can basically only see the details when backlighting.
