That was a nice little sound trip you've made, I liked it :hatsoff:

And thanks, that track is actually pretty far away from how I did rap music, but it was just one of those beats I felt compelled to write lyrics to :muppet:

Relatively new to agar. Here is some pan cyan - does this look bacterial/contaminated? Seems like it isn’t a clean plate

bit hard to tell with the color but looks ok to me. maybe ask in the pans thread

Will do, my other plates don’t have the little brown splotches. I think I’ll avoid the pink plates in the future , they are a bit hard to read compared to the boring light brown ones

Thanks! And the plates are Vabiooth autoclavable plastic plates from the 'zon. Lots of people seem to use them around here. I'm liking them a lot so far.

Check them out

Thanks for the link
I'm going to try them.

I saw a YouTube video of a guy using the water from a grain boil to mix with agar to make plates, is that a pretty normal practice? Seems to work very well for him

Grain water agar is a thing, and lots of folks use it.

I have two problems with it. You never know how nutritious it is, and there is a possibility for toxins/antifungals that some grain is treated with to build up in your grain water and end up in your agar. MEA is so easy and cheap I can’t justify using grain water. I do pour it on my compost pile.

I wanted to know just how nutritious grain water is so i made grain as normal, measuring out just enough water to hydrate a brick of coir, sterilized some coir hydrated with it.

I then injected 20ml of the same lc into 4 seperate bags. 2 bags were 3 lbs straight grain water hydrated coir, 2 bags were 2lb coir 1lb hydrated grain.

No cube myc recovered on either grain water bag. As the aio bags were finishing up i checked the other ones, small mold colony on one from an improper bag seal, but otherwise nothing to note.

Hardly a conclusive sample size, and im sure theres something in there to be utilized, but im still not sure how useful it really is.

My wife takes every bit of grain water I have after hydrating my oats for her house plants. They love it!

I don't like the scalloping.

If you have other transfers I would skip this one.  Otherwise run it out on water agar or through a cabin.

Hey, first time using agar.

I added a drop or two from a spore syringe and this is what i have now. I only have 4 more agar plates at the moment and was wondering what my best option would be to clean this up?

Shoukd i take multiple samples for my second generation or since im a bit limited on plates, just take one and try to get another 2 or 3 generation from that?

You are correct. And getting them with a phone is quick & fast. Maybe one day in the future I’ll experiment with my real camera.

if you use manual focus sometimes you can get it to focus on the culture instead of the lid but its real fuckin hard.

I'm testing LC of psilocybe azurescens that I bought online. 4 out of 6 dishes looks promising, can you please help me with deciding what should I transfer further? The plates were inoculated 13 days ago.

Plate 1 has malt (not malt extract) agar and mycelium looks very thick:

Is such a thick mycelium a good sign? Or rather a sign of some contamination and mycelium gets thick to fight it?

The plates 2,3,4 are grain oat water, with a little of activated carbon (200mg to 500ml), added mostly for colouring. I can see there some contamination (different colour). Are all the other white things mycelium?

On plate 3, 2 colonies joined together, that's why it's uneven.

Hello,

A question about storing plates.

I've done some pour 12h ago, i've let them in the SAB during this time (did not flip those had to go).

1. How long should I leave those in the SAB ? Can I remove those immediatly after pouring ?

2. If I don't plan to use those, do I wrap them ? If yes, how many hours after the pour ? When no condensation anymore ?

3. If condensation dropplets fell on the agar, is it bothersome ?

Ty

Let them.cool until the agar has gelled , couple hours is more than enough, wrap them until you're ready
