agar plates...

it has been sorta cold for these, I am making a TnT, just waiting for the aquarium heater from ebay. So is this normal, these are malt agar plates.

the I would have to say they may be contaminated, the spread is far to uneven
unless you inoculated it really odd.

were you using a flowhood or a glovebox

like i said ive done very little work with agar. priz, is it best to shoot agar with a syringe or just scrape the spores directly from the print onto it?

it's better in my opinion to use a print, the excess water from the syringe increases the odds of contamination.... if your doing it from a syringe as an only option you can remove a lot of water by standing it on it's plunger for several hours and then evacuate most of the water by pushing down on the barrel


to inoculate the plate push a drop out and drag it across the surface, make sure you squirt a little first to get spores into the needle

there are a couple of method for doing it... I heat my loop, streak the agar
to cool it and make the wire sticky, run it across the print and then wipe a few
spores in several areas on the plate... the fast clean healthy spots get moved to
other plates

Hum ok, I just made these in a rubbermaid container with the lid off not breathing and a crapload of alcohol and lysol was sprayed. I had gloves and fresh clothes on. I just made my glovebox today from the container. I squirted some sryinge spores on these and was hoping for the best. It has been almost a week, but it has been so cold so I don't know, maybe its slow growth? I thought myco was only white but I am reading there is white bacteria?

I forgot to add that I finished my syringe on this so there was a little extra fuild so it was spread all over the place. I looked at it closey and it looks like mycillum

I think squirting was you're problem here, I guess you've read the thread and
have picked up a couple tricks... you can turn your rubbermaid into a glove box
for about $10, dryer vent and rubber gloves, the proper application of duct tape
and a piece of plexiglass, you'll be set then... arag is pretty fun to work with
once you have the environment for it
