i am making about 10 agar plates and i was wondering if i could pour the agar in the plates and then sterilize the plates or do i have do sterilize the agar first then pour it in the sterilized plates?

everything i read says put it all in one container, sterilize and then pour i would like to know if i can pour then sterilize the petri dishes?

I always sterilize the agar and then pour it into pre-sterilized dishes.

how would i go about pouring the agar and then sterilizing the petri dishes? could i put the dishes in a ziplock bag and then put them in the pc?

Sterilize the agar in PC , then pour . The results of doing it in the opposite could be quite disatorous . Plastic petris will melt (I assume you are using plastic) in PC . You also risk not getting mixed up enough , thus not gelling properly .

hey mickey, how can someone pre-sterilize the petri dishes if those are made of plastic? or is it okee withous sterilization, i mean plastic is a dead thing, no bacteria can live on it, right?

A scalpel is a dead thing also, but do you sterilize it before use or not?
The plastic petri dishes come presterilized, so if they are originaly sealed, there?s no need to sterilize them.

yeap. my petri dishes from Mycomania were in a sealed bag. once opened, bacteria are starting to attack, right? the bag has 20 petri dishes, so wtf? 
also my dear greek-speaking Anno you didnt tell me how to sterilize them if needed. does this mean that after their 1st use, they have to be thrown away?

Mine to came sterile in a sealed plastic sleeve . Contams were no problem , but I used a make shift tent with sterile cloth , surgeons mask , lot of pinesol and clorox spray , sterile gloves . Basically just create as sterile an enviro that you can . Open dishes only when pouring , and open as little as needed to make pour . I poured ten dishes , innoced four , had zero contams(with exception of one on purpose contam dish ) , did my transfers after selection with no contam probs . Good luck Yes throw them away after use , if you cant do that , best method to clean them up is with alcohol 90% or better . But why risk contams in future , petris are cheap , unless you use glass which are very costly , these are the only ytpe you can "safely" reuse .

You don't, they should be sterile when you get them. I use glass petris now.

It's not that anything would be living in the plastic. If you opened the petri dishes and let contaminants fall into them, you would have problems. Since I use glass dishes, after I clean them out and dry them, I PC them to sterilize them.

okee. is there a good internet based supplier of glass petri dishes. once i saw my plastic ones, i knew i couldnt fall in love with them; i think i need glass ones.

I think for you it would be better to find a company in Europe. Shipping glass from any company in the USA to Greece would be expensive. Glass is heavy.

Glass petri dishes cost around 2 euro per piece here. Shipping for 10 dishes would be around 6 euro.

I managed to get 70 pyrex 60mm petris for 35 dollars off ebay. They have the setup for any type of mad scientist shit you might be interested in. And as far as I know, prepouring agar before sterilizing isn't a problem if you're carefle. (never done it myself, so I can't make promises)

its messy. remember you can sterilize plastic or glass petris (and just about anything else on the planet) with ALCOHOL. its generally available everywhere, cheap, not messy and effective. i guess the only draw-back is that it is highly flammable and therefore kids should not be messing with it, particularly when they have their kitchen stoves on, doing oven teks, scalpel flame sterilizing or what not.

thanks for the help everyone, i decided to just sterilize then pour, been two days since i put a drop from my syringe on each one, hopefully ill see something in the next week.
