yea going to do transfers from other plates this morning but, if i wanna save them for later, which i do frequently,  ill just wrap them up in some plastic wrap.

just leaving them stacked like this overnight the condensation is gone from the lids for the most part. there is still droplets on the walls but they evaporate after a couple days.

ill post a couple pics when i finally get around to transferring

edit: u can see where the warm agar was sitting on the top and helped get rid of the condensation. i usually use liter size bottles that cover the stacks better

Nice! What are your plans with them? Plan to use them soon? I'm an agar noob and have been dealing with alot of condensation in my plates despite pouring at <120 deg F.



I was having so much of the same problem, until I started thinking about how condensation is tied to ambient temperature. Pouring cooler doesn't make it entirely go away, and puts you at risk of chunks. So my solution that has worked wonders for the last couple rounds of plates I poured was that when they are sitting in front of the hood cooling, I put a table at about the same height 3' away from the hood with a *infrared/radiant* space heater on it, which heats the surrounding air *and* helps the plates cool more slowly. Result: crystal clear plates and not having to raise the entire room temp to 90.

Or you could just put a flat bottom mug of hot water on them lol

I tend to just let em air out before sealing em up

:whathesaid: I tend to just do plates well in advance but if I need a plate and it has condensation I always keep old plates handy so I can stack and use an old plate on top to pour hot water in

The problem with hot water after everything has cooled is it's temporary, condensation just comes back a few hours later which is annoying. I poured at 47C (using a sous vide water bath), put a flat-bottom glass of hot water on the top of each stack, and leave them to cool in front of the flow hood for between 8-24 hours. Couldn't get rid of the condensation, which is why I finally went down the path of heating the area. But I work in a larger room that's pretty hard to heat so going with an infrared space heater right on the plates finally solved it. 2-3 hours cooling in front of the hood with the space heater and 100% of the plates were crystal clear.

It should be noted that it's summer, and humidity is regularly 90% outdoors, I work in a semi-conditioned and imperfectly sealed space in my basement where humidity can be 60+%, and I air condition to 68 degrees, so it's cool but moist. I suspect this is the worst possible condition to pour plates. We'll see if these shenanigans are necessary in the dead of winter when I have to humidify just to get it up to 25%.

Another simple fix is to simply keep some sterile swabs handy , just soak that shit up if it bothers you but whatever works and gets the job done

Sterile swabs to sop up condensation is brilliant! Can I throw some non-sterile swabs in a jar and PC with agar to sterilize?

Nothing wrong with condensation.  It can and should be resorbed into the medium.  I go ahead and use the plates with condensate, and it clears up.  More humidity is good®.

When stacking your plates after a pour, keep in mind that nasties are falling from above all the time, especially if you are in the kitchen.  I move my stacks into the lab next to the SAB and cover with clingwrap until use.

When pouring, only tip the lid up far enough to pour, then close. 

When culturing only tip the lid up far enough to work then close, even in the SAB.  I do this -and- culture my plates as vertical as I can,  and then close with my thumb. Have had virtually no contam since these (and other) practices, with six species.

I am always surprised to see those mixing 1.5%, 2%, and more MEA.  I guess my one post on this has had no effect.  You want lower nutes for contam to eat, and you want the myc to be reaching out for nutes so you can ID strong fuzzy myc.

At first I had horrible mold problems with my MiB species.  I found that mold often puts out long strands reaching out and over as much as 25mm before landing again on agar.  See that, you have mold.  Do a transfer the far side from it immediately and cabin.

Short answer- yes

but I usually foil wrap bundles of 5 to preserve sterilization in what I don't use immediately after opening. I also sterilize foil squares for taking prints and store a few piece in a plastic baggie. I know it doesn't make spore printing sterile but at least I have eliminated one contamination vector.
