I've had so much problem with contamination despite all my efforts (above), that I am hedging my bets.  Also it gives more opportunity to select for different types of growth.  Four plates in one.  I take a pic of each plate and put it in my spreadsheet to its numbered entry.

Also I can hold the plate in one hand, slide the lid over just enough to set the slice, then slide it closed with my thumb reducing contamination.  I am now holding plates vertical for most opening operations.

I can send a quadrant to each quart jar and track the results.  Gotta send the MiB soon now.

Lab (bedroom) is right next to the bathroom which can't be helped, but I'm keeping the door closed.

Understand.  Notice I have with some.  But most ontamination is elsewhere on the plate.  Operating vertical from now on for me.

What, no MiB 6?  Well it was too sick, so took what I could to start MiB 16.
               

Yep, I have excised all the green out of the plates (right after these pics), and it should clear up alot.

Have a look at the War when MiB 4 met trich... looks like it wants to form a plin, although I have cut that and the trich out now.  Curious that MiB 5 grows so much slower... these were made from plins.  It'll be interesting to see how each develops on grain.

Huautla 9 and Ochra 13 were taken after the excisions.  1% is close to jelly-like and it is not easy to get excisions out before they fall apart.  Sure can't tweeze them.

You're going about this backwards... You don't remove the bad from the good. You remove good and put it on new plate.

Ok.  But my reasoning is that the green is such a small surface area compared with the good, that it is sensible to remove the bad.  In either case if the bad has sporullated the good is contaminated.

With as jelly-like as this 1% media is it's not possible to lift a whole plate out in one piece withot folding/cross-innocing/breaking/getting air dust all over it.

I considered your input.  We also seem to have different ideas on the nature of myc we select for: mine is fuzzy, yours is rhizo.  This is the nature of science.  The nature of Life.

It appears to me that almost all the action happening at 09:00 is green?
Also, has anyone seen the yellow stuff at 06:00?



Looks like I missed a copious spot at 10:00 that must be cut out right away?



This to me is amazing.  It is thickening to try and extract more scarce nutrients, and is expanding beyond the desert of plastic to find a source.  My grain isn't sterilized yet!

With love, bro just transfer to a clean plate. 

A lot of the contams on your plate are invisible to the naked eye until they start growing out.  If you're just excising the visible contam you still have a dirty plate as the entire thing is covered in things you can't see yet.

Edit:  I now see that there is an unidentified cloud covering half of the first and second plates.  Bad news. 

The yellow puke on the third at 06:00 can't be good either, but the fact the myc joined without fighting indicates they're the same pheno.

Sure glad I now know about water agar.  Should have gone to this before 1% MEA.

Hello everyone!
I'm a noob wanting to get into agar. I bought a pack of ten glass petri dishes which I pc cooked and poured in my SAB (Though I need to work on my movement in there). Sadly, all of those dishes got contaminated by bacteria, so now I have made pre-poured plates that are cooling after a session in the PC. My question is how I should go about taking the plates out? The PC is to big for my SAB. Is opening the PC in the open room and quickly sealing each plate with cling wrap a decent idea?

(Oh and I've tried with plastic plates too but they stress me out much more for some reason)

sounds like your only option. gonna have to open and wrap the plates as is. i would just advise against doing it "quickly".

and hope you didn't get a bunch of boil over out the plates.

You might be looking at another do-over depending on how things go.
Try stacking the plates and wrapping the stack in foil before PC.
Then you can unwrap them in the SAB, and it’s no issue to open the PC.

Edit - I don’t do pre-pours though.  I pour agar into clean plates in the SAB.

just make regular plates. you're getting way too fancy and it's biting you in the ass.

mix your DME and agar powder at 1:1 ratio. add powdered mixture to water at a rate of 20g per 500ml of water. and sterilize solution for 30 min.  the math is easy to extrapolate up or down. make what you need.

stop putting multiple transfers around the edge of the plates. put one transfer in the center of each new plate. take transfers of good looking myc and transfer to new plates to isolate away from contamination. don't cut contamination from plates and then leave the plates to further grow in hopes of transfer.

i don't know how much clearer to make it. you're getting good myc growth but you're dropping the ball by having your cart before your horse.

i hope that helps. and if it doesn't.... maybe try growing basil?
