You're welcome.

x7x (without the plus) is not as robust of a grower, and not as strong. Link on that in my sig line.

this plate looks like it has some ryzo area around 7:30-8 o'clock close to the centre.  It disappeared though as it grew out.  Can I take a sample from the ryzo area and make transfer?  what would have overgrown the ryzo section or did it just change up to tomotose??

That thin growth trips me every time. I cant trust it...I still dk if that's even mushroom myc or mold or both.

you mean the rhizo actually receded or, it was overgrown/outpaced by the more fuzzy looking myc? you can def try to isolate it regardless. i really like the south/southeast portion of that plate. looks nice n healthy. maybe transfer a few samples from that plate and see what happens.

I know you were replying to him, but I'll ask ya anyway since you seem to know - all the growth other than the real ropey rhizo- is that typical healthy myc? My hawaiian plates look like that, but the thin, whispy behavior weirds me out. It's basically just like this pic.

Healthy myc can be either rhizo or tomentose. I’ve almost given up trying to get rhizo growth since all my plates look tomentose no matter how many transfer I make. However once I throw them on wheat jars the fuckers start to look stronger and “ropey” so I’m just convinced the issue is my agar and not the culture (that apparently was healthy all along).

Yeah, might try playing with your agar recipe if you want rhizo growth.

Yeah I've been playing with every variable for the last 3 months, I think I narrowed it down to the agar brand or the brf sedimenting to the bottom of the plate after PCing them as no pours. Tomorrow I'm trying a new agar and doing normal pours so it stops sedimenting. Hope something changes, if not I don't mind, fahtster did magic without agar for years

Why increase nutrients? I understand that decreasing nutrients should encourage rhizo growth due to the culture being in "search mode", but what is the issue you want to remedy by increasing nutrients?

I've read this as well and experimented it with over 30 plates... my 2% nutes look waaay better than the 0.5 or 0.75% ones. I don't know if it's because my sedimented plates are already low nutes on the surface, but the myc is almost invisible in the 0.5% nutes. At 2% they are "weak rhizo", not thick ropes but at least I can see the thin strands. Like this one:

I run .5% for tomentose cultures (ones that are incapable of going rhizo. And 1.2% for rhizomorphic ciltures

its hard to say with absolute certainty but that plate looks healthy all around imo, despite the variations. perhaps something goin on on the north side but its too out of focus. I will typically transfer until i see more uniformity and choose the "thicker" sections just bc. most of my cultures are more wispy and tomentose and do great. i dont try to get "rhizo" growth and when i do, those cultures dont necesarily do better. its definitely seems variety dependent which type of growth. ive been using the same agar recipe for years so i cant comment on that but sediment or not, doesnt seem to affect my cultures.

all these plates are healthy and show some serious wisp..

Thanks for the info man - that pretty well clears it up. Probably 75+% of my grows have been nats, which are ropey af pretty much all the time, so I've kinda struggled with the cube myc a bit. I wasn't confident that that thinner whispy growth was healthy myc.

Okay I have to beg some wisdom.

My questions boil down to:

1) What nutrient and agar levels are good for germ plates in particular? I've been making 2% each so far but I made some grain water plates and I think they're way lower nutrient.

2) How do you estimate grain water nutrition anyway? The recipe I looked at said something like 2/5 grain water to distilled water. The pictured plate is 1/3. I made a second batch with a little more but haven't used yet. Going to use those for the upcoming transfers. The grain water was from boiling oats and should be about as concentrated as grain water get but the growth is really underwhelming compared to the left over MEA.

3) In germ plates, is there a clear indication for whether or not the spores did their thing or if a colony is from a solo spore and will be non-viable? I shook my syringes with everything short of lab grade automation, gave the plates a solid 5+ drops of fluid then sloshed it over the plate, and got something growing on every plate but a lot of them are less chaotic than expected. Like, 1-2 fingernail colonies (2 weeks post inoculation) with nothing else on the plate. Did my shaking suck and germinate only one spore or should I transfer these colonies like they're a viable line?

Pictures:

Here's one of those "one thing grew" plates. Pan bispo. No, it's not really any clearer in person. I'm happy that the plate is so clean and growth is uniform but should I expect this to be viable?

Here's some contrast between the MEA and grain water plates. Natal Super (or whatever we're calling this line at this point). The bottom one got some clumped spore chunks and is the most vigorous growth on any of the grain plates. MEA plate looks pretty good too and initially had two clearly distinct germination points.

Left subtrop. Needs cleaning up badly and was a bit more what I was expecting in a germ plate. The colonies are starting to come into contact but mostly seem to be shrinking from each other. Does that mean they're incompatible mating types or are they done mating? I present that one for illustration because some of the grain water plates look similar but are even less photogenic about it. Right is bispo again. That syringe must be nice and clean but again I have two very clean growing colonies and I'm not sure if they're viable or if I need to let them grow into each other and watch for clamp connections. I was expecting so much more chaos and to not have to think about it after putting a minimum of 5 drops on each plate.
