Re: AGAR ENVY! (Anything and All things agar!) [Re: AspectOfTheCreator]

What you guys think of this plate?
Mold?

Can someone explain to me why does this occur?

Looks like a plate I'd transfer from. I think.

I've transferred like 5 times from a plate that looked like this. Then all of them dint looked like this. And suddenly this occurred again...

I’m pretty sure I figured this one out a while ago.

So the next tiem you transfer, pay attention to what side of the wedge was the leading edge of the myc growth, and which sides were from further down. In my case that weird spot was always starting from the side of the wedge that was the edge of the myc growth.

Could just be mine, but I’d bet it’s something similar. This also happens when there’s mold meshed in but it looks a little different. Yours looks all clean to me.

It could also be from something like the wedge not sitting flush on the plate. Like if one side is slightly tilted up off the plate it might take the mycelium a little longer to grow down to the surface before spreading out whereas the rest of it just has to grow outward because it was already touching surface.

Idk why but this used to drive me insane to the point I had to find an answer

Insane you say... I'm dying over here doing my research to why this is happening and what is it...

The new pics looks uniform under the agar where the suspicious area is..

It looks like something is riding along with the wedge you transferred.
Try transferring solely mycelium to a new plate and leave the old agar on the plate.
I did this recently and the new plate was much more uniform/circular growth.
Is your nutrient composition the same on each plate? Different agar batches?

I This transfer came from a very uniform and healthy looking plate.
I put a piece from the healthy looking one on some grain . I will wait for the first shake and see if it goes bad or not.

This agar batch was 6g lme 500ml.
What do you mean diferent nutrient composition?

If I swirl the bottle each pour?
I turn of the stirplate when it's about temp.

Do you remember the direction of the scalpel transferring the piece?
Some people make a habit of flaming their scalpel handle as they believe it can allow contamination to ride along/fall on the new plate while transferring since typically only the blade is sterile.

Ex. The afflicted area in that photo is to the left, good growth to the right, so perhaps hovering the handle over the left side while placing the new piece allowed something undesired to fall onto that side

Taking note of directionality of agar wedges during transfers now.

In other news, whatcha think of this plate (minus the small rant)? I think I've got a new LC candidate? Maybe? In any case it makes me happy to see a nice even circular growth. Yay. Hopefully. :boofbanana:

I flame the tip (gigity) and all on the area that hovers the plate

If it's meshed in mold.. when thrown into grain, what indicators I will have to be on the lookout for. If it's mold,  shouldn't be growing exponentially fast.?

:bow2: What’s the recipe? I just been doing 10g agar, 10g LME, 400 ml water but I think I want to try less LME maybe 7gs.

Day 3 of t1 on these ghost plates other one looks kinda meh so I’ll be focusing on this one… I THINK I might have done something boys & girls I know it’s early but what do we think? Also the ksss ufos are showing zero growth which sucks but at least I know my tek is somewhat clean.

Nvm I think it’s just mold again

Your technique looks clean and the plates certainly look clean at this point.
Just start over with some new spores.
