Any reason why I can't seem to get any rhizomorphic growth? I got Ecuadorian Cube spores, so nothing particularly exotic. I think so far out of about 14-15 BRF jars and lots of plates (admittedly those were much less "successful" until I started doing water agar first) only like 1-2 BRF jars had some rhizo growth among clouds of tomentose mycelia.

I germ all my spore on brf and only seen a couple show any rhizo growth so far. I’ve seen people say lowering nutes in your plates can encourage rhizo growth but it never did for me it just made my growth weak and slow. Any cube culture can switch back and forth between rhizo and tom and back again, I wouldn’t worry about it too much personally.

Don't get hung up on rhizo growth. It's not an indication of something that will be better than tomentose. It's far more important to just have clean cultures. It's possible for a tomentose plate to go rhizo then go back to tomentose. All my cultures are tomentose and they all preform great when I do things right.

Yup, exactly the same thing for me. Did like 15 water agar plates with pieces of a BRF puck and pins from the same puck (pinned invitro) none produced any rhizomorphic growth, only a very thin layer of fluff.

Ok, will work with the stuff I got, chasing rhizo growth didn't seem to yield any results anyway.

I inoculated some plates today by putting one (1) drop of spore solution on the agar and spreading it around with the loop.

But several hours later I was looking at the plates, and I noticed that rather than being absorbed into the agar, the water is still sitting on the surface of the agar and rolls around if I tilt it.

The agar I prepared contained 8g agar-agar and 8g light malt extract dissolved in 400mL water.

Is this a problem, and how do I prevent this?

Thanks. The reason I thought it was an issue was the common admonition against allowing loose water to roll around inside the dish, such as by carelessly squirting spore solution from a syringe, or having too much condensation. The usual reasoning is that the loose water will spread contaminants further.

Yeah don't worry about that. Condensation is just bad for visibility. It can help spread contam if you introduce the contam, especially bacteria they get spread around really nice by water. But if your sterile technique is good then no worries, the condensation is gonna be as sterile as the rest of your plates.

You did use a spore syringe though so contam is almost inevitable but you should still get plenty of myc to transfer away from the contam

Yeah, well, we can hope the plates are sterile :grin:. But the edge of the dish can be a dangerous place.

Well the mold plate grew out a bit after 2 weeks, seems to be a bit less picky than the cubes, still seems a little tame for 2 weeks but it's growth

Got my new agar agar in the mail today, will be making up new plates tomorrow

I would like some input on these plates. Where should I transfer from? There’s one natalensis, which is the spore plate. 3 semperviva, transferred once.

Also I have 4 bags of purchased sterilized rye. I was thinking I’d do 2 semperviva and 2 natalensis. Can I do a Josex poke to nocc them up? Or do I need to make sslc first? They have injection ports, so to use an agar wedge, I’d have to cut them open so if possible I’d like to avoid that.

My last few germ plates from spore syringes took 3-4 days until visible colonization, about a week for visible mycelium and 3~ weeks for plate coverage

How do you all identify sectoring?

I’m not sure there is a consensus amongst the community and if there is I would like to know how/what everyone thinks.
