But I do know when I see a trend. When I describe consistency in a grow I am not saying consistently good. 9/10 of the grows are failures. But they fail consistently.

What I am saying that in my experience (from multiple spore sources and over the course of many years) that there is a consistent trend in the way they grow.

The mycelium colonizes the substrate the same amount of time. The pinset looks the same. The morphology of the mushroom is the same. The the flushes recover in the same time. The product weight is the same. Of course not exactly the same, as there are always variable. But down to less than an ounce of difference.

And through all of my time growing, it is the same example that I gave no matter the spore source. If I inoculate a Petri dish MS, and take 10 transfers every single grow will produce something different in morphology, product weight, colonization times, etc.

That inconsistency doesn’t happen with T100. It might be inconsistently bad, but consistent nonetheless.

Why is that? (Again, genuine question)



I ment the same consistency you speak of. I wasn't talking about success or failure. And I and others have answered why this is. Not sure how to give ya more answers than all we have.

If you're getting spores that show all sorts of inconsistency than that's actually a good thing really. It's great for pheno hunting and stabilizing whatever you want. Variety and vigor is a good thing!

Doing lots of transfers and getting more consistent result variables is also good! Just ur way takes a long time and requires then preserving that culture forever.

But the same results can be achieved through generational breeding then you get spores that don't have the extreme variation in results as much.

I understand you. Maybe it is that my goal is to not work with spores. I don’t collect spore prints. I really like to be able to grab a culture out of the fridge and know exactly what I’m going to get. And it has worked for me…for now. I don’t get the same consistency on transfers from the same dish early on.

I’m also not trying to breed anything, which was my understanding for the need to collect a dikaryon produced from two monos for a “pure” strain.

I just want to be able to throw some myc on a plate and know how it is going to perform.

Then it sounds like you're achieving your goals. Just remember it's not a true isolate. I personally would use cultures too fast to store them in that way. I prefer generational breeding so that it doesn't matter if I'm using spores or clones it all pretty much looks and preforms the same with only slight variations.

If I ever find my genetics are getting weak or losing vigor then I'll follow the advice I've been given to then cross that with another variety. Along the way if a certain clone is awesome then I'll store a bunch of it in distilled water in cryovials.

The OMC has been misusing mycology terminology for a long time. An "isolate" is an isolated strain. A strain is either a single dikaryotic fruiting strain, or a monspore non-fruiting strain. Stro uses the terminology incorrectly in his thread on cleaning cultures.

It's possible that monosporic isolates are not considered actual strains, however, mated monosporic isolates do make a strain. This is incontrovertible. Monospores are certainly isolates though.

It is my understanding that a strain is defined by the sexual reproduction of any compatible hyphae.

Please clarify so I don’t misuse it here. You are saying that a strain is only produced by two monokaryotic mycelium?

Yes. So, an isolate is an individual strain. When the mycelium branches and grows the likelihood of selecting an isolate with your scalpel, or even a needle biopsy, is impossible.



Individual strains are microscopic. Any attempt to transfer even an imperceptibly small amount of material will still contain many strains.

Okay so strains are constantly being reproduced. So they are strains, you are just saying they are not possible to isolate?

every small transfer is limiting the number of strains. Until eventually it is isolated.

What I gather reading online is that an isolate can refer to an individual or a group of individuals that work separately from others within the species.

So maybe my confusion is how you are using the term isolate in “isolated strain”. Do you mean to say that unless you separate two spores and collect their dikaryon offspring, that it is not possible to collect an isolated strain?

If you could isolate single straina on agar, labs wouldn't employ tedious techniques like serial dilution or dedikaryotization to produce individual strains.

Is that a factor of time? I spent a year transferring plates to get something like 8 worthy strains. If I had lab equipment I wouldn’t do it this way either.

Everything I have read other than on this website says a strain is not limited to two monokaryotic donors.

Which sites? I'd be interested if the information is from a reliable source. I have been reading mycology papers and books for some time now, it would surprise me if I missed something as basic as this but anything is possible. I'm not infallible.
