Hi everyone, got a question about maintaining a stock of myc on plates in the fridge.

Is this a good idea? Or do you guys start from spores usually? The ones who don't have a grow operation running all the time.

I keep thinking it would be nice to isolate a vigorous myc at e.g. T4 and put a couple of plates in the fridge to use later.

The question is how do you maintain. Do I do a new transfer in, I don't know, 2 months, 3 months or something of the sort, and then put that in the fridge to continue maintaining or how would I go about doing this?

The idea being if I wanna go to grain I do a transfer from the fridge culture and once it covers the plate go to grain, thus skipping germination and multiple transfers to get a clean and vigorous myc.

Yeah, that'll work. Myc should be good until the agar dries up and a couple months past really. It's just a bitch to get a transfer piece from agar that's like a potato chip but the myc will still grow if you can manage to get the transfer piece

So proud! My spore print to agar Experiment was a success! I’ve got 1 of 8 that didn’t take but I can live with that. This hobby is really starting to stick. I’m loving it. Thanks to everyone that has provided info so far this place is great!

I know the pictures shit but I’m proud. Is there anyway to deal with the condensation if you failed to mitigate it when you poured the agar cups?

Try and get a pic with some backlighting.
I've been using deli cups recently, without micropore, and the condensation dissipates within a week. Just carefully pour it out before making transfers or you can open and work with them upside down, keeping condensation in the lid.

Hey all. I'm really inexperienced. I had to prep my agar a day before I finished building my flow hood, and when I warmed it up in the microwave, it got to boiling but when I poured, there were several fairly big lumps in the agar. Is this normal? Should I be holding the agar bottle at temp for a while so this doesn't happen?

Hi, I don't think noobs like me are supposed to give advice, but I guess someone's gonna say something if this is no bueno.

I do it in a pan on a stove and use a nice wisk to have everything homogenize nicely and the I test the temp so it's between 120 and 140 F and then pour the plates (I do no pour).
Sort of treat it like making a fine meal. Microwave is not high cousine.

The ones that pour in the sab or in front of the flow hood would either liqufy it, pour it in a bottle, pc the bottle and pour when it gets to around 120-140 F.
Or put the solid agar that is in its bottle/container straight into the pc and then pour.

For the no pour it's no biggie, I read some of the experienced cultivators put a solid chunk of it direkt onto the plates and then pc the plates.

Haven't come so far across a tek or a post from a trusted cultivator who used a microwave.
I'm sure there must be a ninja on here who nukes it to perfection though.

One question for you. You say, really inexperienced, but you registered 2008. I repeat, 2008! How that happen?

ive melted agar bottles in the microwave countless times. i usually PC 5 or six 500ml agar bottles at a time but i dont pour em all. so i back the cap off and melt that shit on medium for a few minutes.

i wouldnt use a microwave to sterilize agar thats dumb.

It's normal, I like to pour my plates really cold so sometimes I get chunks or bubbles in the last couple of the batch. Mycelium will eat it and it doesn't attract contam.

I make agar in the microwave sometimes to test recipes without busting out the PC. Then when I want to pour I'll PC an hour beforehand. Throw the bottle in a water bath around 120 degrees. It will heat the water up, then the whole thing will cool down to 120 and you can pour.

Bod just uses it to do the initial mix and then PCs it. I don’t really see much of an advantage though. Just mix it up the stove.

Ummm.... If you're sterilizing agar in a media bottle in the pc there is no reason to pre-boil. Just throw everything in the bottle and run the pc. No need to boil it so can you boil it.

Per-boiling is only done for no pour(should be called pre-pour) since you're pouring it into individual containers before sterilizing.

Get it together guys. Don't make me get Stipe in here with the belt.

Homemade malt extract before PC:


After PC (sad to have gotten so much condensation on such clear media)


Corn starch and brown sugar media:


Hopefully my sterile technique was up to game when inoculating a bunch of them πŸŽ‰

I’m using deli cups as well, as mentioned upthread I also make a few bottles and store them in the fridge..

I would say next go around try and pour as cool as possible.
After I’ve brought my 250ml bottles to temp in the microwave, I let them sit at room temp for almost an hour..almost to the point were it starts to congeal back up on me after I’ve poured about 2dz cups. Almost knocks out condensation completely IME.

Hindsight has a tek for little-to-no-condensation. It looks like too much work initially but once you get into it, you wont go back.

100% condensation free agar plates pour or no-pour

Promise to add a pic next time I post in this thread.

Yeah, I had read that one. Tried it out with a previous batch and made a mess lol. Don't have enough free kitchentop space for it to be worthwhile I think. Also my wife has been tolerating most of the weird shit I've been doing for this new hobby but the duvet on the kitchen top was a bit much haha.

Thanks anyway :laugh:

I now wonder if just putting them in the oven at a lowish temp and only the top heat on would work. Might make things worse lol.

I disagree about not requiring shaking, maybe it works for you but everytime I just poured in agar and pced i got a puck on top and that was just .2% agar for sslc. Ive always had to homogenize the mixture first
