So you accidently dropped a piece on the side, then decided to put another one in the middle, which you failed to do again?

Lmfao yes, great use of Occam's razor. Definitely more likely than I sliced the sample in two when getting it off my scalpel and one piece slid over

Maybe don’t let Occam do your agar work anymore then.

Quick question…I’m new to agar…I was using black agar and getting nowhere so I started leaving out the carbon and things are going much better. When I use back lighting to take pics of my agar…I noticed my agar is β€œgranular”..can’t really see it without the back light…is there a good way to get rid of that? I’m using a simple recipe of distilled water, agar powder, and LME

You could boil some water and put the lme in it and stir, then use coffee filter and add to your media bottle with your agar, then PC...

Probably don't filter the agar...

If you use a dash of nutrional yeast it also leaves a little residue that you can filter out...

I use 250ml bottle with 210ml water and 3.5gm LME, 3.5gm agar and .5 gram filtered nutrional yeast broth and my cups come out pretty clear'ish...

Awesome that sounds like a good idea! I have coffee filters already and a cone holder..I will give that a shit next batch…doesn’t seem to affect it working right or not but I don’t like seeing it grainy lol

An immersion stick blender when it's hot on the stove works great to get everything dissolved

Nice…I don’t have an immersion blender right now but if the coffee filter trick doesn’t help I will look at getting one

I just heat up my Agar on the stove and use a whisk. NN for anything more, just don't boil it. Just whisk it until a lot of steam is coming up. You will also notice a slight foam that builds up, those are the nutrients from the LME being dissolved and it will get clearer when the Agar is all dissolved. But never boil it, Agar needs ~85Β°C to dissolve.

That's how I do it and never have like "flakes" or "granulat" in my Agar. I also don't filter it.

(edit: you actually don't even need distilled water, normal tap water does just fine)

yeah no need to use an immersion blender unless u have one already
I heat on high and whisk until its about to boil over then take it off the stove
and use filtered water if you can - that got my plates really clear

edit: I've stopped using food coloring all together its just an extra step I don't find necessary

I only use 3 unbleached filters for the NY, but I guess if I used 4 maybe it would come out clearer!?

I haven't tried making both the LME and NY filtered broth yet cause my cups really are tasty to my myc...

granular is fine. thats how mine come out. why stress over something that has no effect on performance? but, i will say this, just keep an eye on sediment in agar to make sure its not "expanding". contams can pop up inside the agar layer whether theres sediment or not but, sediment can make it a lil more difficult to spot. i recommend always paying attention to the whole dish when checking for growth. dont get tunnel vision on just your transfer myc.

mine are even more granular now since i decided to give nutritional yeast a try to see if i notice a difference in performance.  the jury is still out on it.

i used dye for a lil bit back in the day but unless i'm making different agar recipes that i want to easily tell apart then, i'll use dye. but, i like to keep shit simple whenever possible. 50/50 blend of malt extract and agar mixed 40 g to 1 L of water and sterilized for 30 min.  new addition, about 2 g NT per liter of solution.

i've tried both filtering and not filtering the solution. my current preferred method to get the most uniform sediment dispersion is by sterilizing the the agar solution with a mag stir bar added. that way i can stir the sterilized solution. we'll see how long i keep doing it this way.

i also dont wait for any certain temp before i start pouring. i pour hot and use the dishes with the least condensation first. the rest get put away and any condensation is usually gone after a little time.

This hobby can get complicated easily.  and it's also easily over complicated.

but hey, what do i know?

It really doesn't matter if you boil it - as long as you pour into media bottle before it solidifies to avoid the hassle. Your PC will just reboil it anyways
